Objective:To study the intervention effect of different extraction from Chrysanthemum indicum L(CI) on eczema-lesion like mice, discuss its pharmacodynamics and probable immune mechanisms.Methods:56 clear KM mice were divided randomly into 7 groups named blank group, model group, positive drug group(Erfukang liniment), aqueous extract, 95% ethanol, 70% ethanol and 50% ethanol of CI group. Eczema lesions mice model (except blank group) were established by 2.4-dinitrochlorobenzene (DNCB) sensitizing-challenge while all drugs were given after initial sensitization(dl).All the animals were sacrificed in the day after the last challenged, the swelling, pathological changes of challenged right ear by hematoxylin and eosin(HE) were measured, and the serum of all the mice were collected. Then fingerprint of CI distinct extraction were established by High Performance Liquid Chromatography (HPLC) to choose the best two extraction group (more kinds and quantity compound composition existed). Finally in the five groups the serum IgE were tested by enzyme-linked immunosorbent assay(ELISA)and Immunohistochemical method was used to measure the level of Interleukin-4(IL-4) and Interferon-y(IFN-y) in right ears.Results:1?Histopathology change of right ear from the model group are consistent with acute eczema skin pathological changes and the swelling could be significantly increased compared with the blank group(p<0.01). Compared with the model group, the swelling get significantly decreased in the another 5 groups.The positive drug group and water extraction of CI group have a obvious difference with all the ethanol extraction groups(p<0.01) while there having no difference between themselves(p>0.05). Pathological changes of lesions can be controlled in all the groups (except blank group) at different degrees;2?Fingerprint of CI distinct extraction are successfully established by HPLC. The particular peak can be found both in aqueous and ethanol extraction of CI.The aqueous group could express more peaks than all the ethanol extraction groups which can be seen the best expression in 70 percent concentration in the same circumstance.3?There is a significant difference between the model group and blank group with the elevated serum IgE and skin IL-4, co-existed within the decreasing expression of IFN-y(p<0.01). Compared to the model group,aqueous extraction can reduce the high IgE level (p<0.05) and particularly achieve decreases in IL-4(p<0.01),stop the decline the expression of IFN-y(p<0.05) that can be significantly seen in the ethanol extraction group(p<0.01).Also the level of IL-4 get brought down (p<0.05) and a trend of gradual decrease (p>0.05) can be observed in the ethanol extraction group.Conclusion:1?Chrysanthemum indicum L (CI) has a therapeutic effect on eczema-lesion like mice, the major medicinal composition might exist in the aqueous and 70% ethanol extract from CI;2?It can be seen as a part of immune mechanisms that extracts from CI can correct the relationship of TH1/TH2 immunity imbalance in T-lymphocyte by decrease the expression of IL-4 to increase the secretion of IFN-y in the eczema.
