Alcoholic fatter liver (AFL) is a chronic liver disease with excessive alcohol consumption, liver steatosis, liver cell injury and inflammatory cell infiltration. AFL is the most important cause of cirrhosis, which is one of the top ten common causes of death. In recent years, the incidence of liver damage caused by the alcohol is increasing year by year in china. Alcohol has become the second leading cause of liver damage after viral hepatitis. However, for a long time, studies have failed to clearly define the molecular theory and the optimal treatment has not been established domestic. Thus, it is very important to study on the pathogenesis and efficiency medicine of AFL .Chrysanthemum is bitter, acrid, slightly cold with the effect of evacuation of wind-heat, swelling and detoxification, anti-infective and anti-viral. Chrysanthemum is widely used clinically to prevent influenza, meningococcal meningitis and other acute and chronic infectious diseases, drugs for the treatment of carbuncle swollen boils, hypertension, hyperlipidemia and cancer, etc. Chrysanthemum flavonoids (Total Flavonoids Chrysanthemum indicum,TFC) is a active ingredient extracted from traditional Chinese medicine, which is used to anti-inflammatory, analgesic and immunomodulatory.The aim of the present study is to explore the therapeutical effects of TFC on AFL and to analyze the possible mechanisma of TFC effects in rats fed with high fat diet and sucrose. The main contents are divided into some sections as follows:1.Protective Effect of TFC on alcoholic fatty liverThe rats of the model group were fed with alcohol and sucrose. The rats of TFC group(84,168,336mg/kg) and tiopronin group (60mg/kg) were administrated with related dose of medidine by the end of the 3th week and for six weeks. TFC effectively reduced the serum ALT, AST. The histopathological analysis showed that TFC can significantly alleviate the symptoms in the hepatopathy rats including hepatic steatosis, lobular inflammatory cells infiltration and necrosis. These results suggested that TFC has positively protective effects on alcohol and sucrose induced rat fatty liver.2.The effects of TFC on alcohol metabolism of alcoholic fatty liverTFC effectively reduced the serum ADH levels compared with model group. . These results suggested the effects of TFC on alcoholic fatty liver might be associated with regulating alcohol metabolism and reducing fat deposition.3.The effects of TFC on lipid metabolism of alcoholic fatty liverTFC effectively reduced the serum TC, TG and FFA levels. These results suggested the effects of TFC on alcoholic fatty liver might be associated with regulating lipid metabolism and reducing fat deposition. 4.The effects of TFC on oxidization of alcoholic fatty liverCompared with the model group, the hepatic MDA was decreased significantly in administration of TFC, meanwhile the activity of SOD was enhanced. These results showed the effects of TFC on alcoholic fatty liver might be associated its antioxidant5.The effects of TFC on inflammatory reaction of alcoholic fatty liverThe levels of TNF-αin serum was remarkably reduced, TFC effectively depressed abnormally high expression of TNF-αmRNA, MCP-1 mRNA and NF-Κbp65mRNA in hepatic tissue. These analyses suggested TFC could down-regulate the cytokine and relieve inflammatory reaction to prevent the liver.To sum up, TFC has positively therapeutic effects on experimental alcoholic fatty liver model. The results suggested that its mechanism of action might be associated with the ability to regulate alcohol metabolism, decreasing lipid peroxidation, downregulating, ameliorating inflammation in hepatocytes.
