Objective:The study is based on the results of the previous study on the inhibitory effect of water extract of chrysanthemum indicum L.and ethanol extract of different concentrations on eczema model mouse skin in different degrees.To explore the Effective material basis of water extract of chrysanthemum indicum L.and ethanol extract of different concentration on eczema animal model,and to isolate and purify its extract in order to find the effective chemical constituents which have inhibitory effect on eczema.Methods:The flower buds were dried with distilled water,95%ethanol,70%ethanol and 50%ethanol for reflux extraction,then concentrated extract and freeze drying machine were used to obtain the solid standard extract of wild chrysanthemum extract with macroporous resin and microporous resin.Reverse silica gel(ODS),normalphase silica gel,porous gel adsorption resin(MCIGELCHP20)and hydroxypropyl dextran gel(Sephadex LH-20)were used to separate and purify the water extract of chrysanthemum indicum L,and then hplc,nmr,ei-ms,was used to separate and purify the water extract of Chrysanthemum indicum L.HPLC?NMR?EI-MS?ESI-MS/HR-ESI-MS?UV?IR et al.Methods 15 high purity compounds(No.1-15)were analyzed and identified and their structures were determined.These 15 compounds were used in the in vitro inflammatory model experiment of Raw264.7 macrophages induced by lipopolysaccharide(lps).The changes of(no)production rate of nitric oxide were observed.Results:1.The extract of Chrysanthemum indicum L.from water and 95%,70%,50%ethanol solution can inhibit the local skin damage and immune inflammation reaction in mice at acute stage of eczema.2.Fifteen compounds were isolated and purified from different extraction methods of Chrysanthemum indicum L..One of them was a novel glucoside sesquiterpenoid,and the other 14 were known compounds,of which 8 compounds(Nos.3,4,8)for the first time,it was isolated from wild chrysanthemum.3.The results of cell activity test of 15 compounds in vitro showed that three of them(No.2,No.3,No.7)showed different inhibitory effects on no production rate of raw264.7 cells at a certain concentration.It can be concluded that the three compounds all have different degree of anti-inflammatory activity.Conclusion:1.Chrysanthemum indicum L.has therapeutic effect on eczema and its effective substances exist in four kinds of extracts.Fifteen compounds were further isolated and identified.2.Compound 2,3 and 7 isolated from the extract of Chrysanthemum indicum L.were effective substances for anti-inflammatory treatment of eczema.
