Chrysanthemum indicum var.aromaticum is a precious plant unique to Shennongjia in Hubei Province.Because it grows in high-altitude areas with abundant sunlight,the whole plant is very aromatic.At present,the research on Chrysanthemum indicum var.aromaticum in China mainly focuses on the essential oil of whole plants or flowers.The chemical composition of the stalk of the Chrysanthemum indicum var.aromaticum was rarely studied.In this paper,the extraction process of total flavonoids from the stalks of Chrysanthemum indicum var.aromaticum,the analysis of fat-soluble chemical components and the antioxidant experiments in different polar parts were studied to study the stalks of Chrysanthemum indicum var.aromaticum.In this paper,the response surface method was used to study the extraction process of total flavonoids from the stalk of Chrysanthemum indicum var.aromaticum.The four factors of ethanol concentration?extraction time?extraction temperature and solid-liquid ratio were selected as the independent variables,and the flavonoid extraction amount was used as the response value.The influence of various factors on the extraction rate of total flavonoids was explored by establishing four-factor and three-level orthogonal experimental design.The results showed that when the extraction time was101.16min,the extraction temperature was 79.69?,the ethanol concentration was63.38%,and the solid-liquid ratio was 21.76:1,the extraction rate of total flavonoids of Chrysanthemum indicum var.aromaticum was up to 2.82%.The original extract of Chrysanthemum indicum var.aromaticum stalk was extracted with ethanol,and a sample of seven fractions was obtained through a silica gel chromatography column.A total of 101 compounds were identified by chemical analysis of seven fractions by gas chromatography-mass spectrometry?HS-GC/MS?.There are 31kinds of terpenes and their oxygen-containing derivatives:10 kinds of terpenoids,17kinds of sterols,1 kind of furfural compound;21 kinds of aliphatic compounds:6 kinds of ester compounds,acid compounds 15 kinds.A high-performance liquid chromatography?HPLC?detection technique was used to conduct a standard control test on ethyl acetate extracts.Three components of luteolin,apigenin and luteolin were identified,and the simultaneous determination of these three flavonoids was established method.Luteolin was measured at 0.372 mg/g,apigenin was 0.0682 mg/g,and luteolin was 0.462 mg/g.The best extraction technology for Chrysanthemum indicum var.aromaticum stems original extract,and,in turn,ethyl acetate was extracted with petroleum ether chloroform n-butanol extraction aqueous solution of the original concrete respectively,finally get the petroleum ether extract chloroform extract ethyl acetate extract of n-butyl alcohol extract ABTS method by DPPH method,Fe3+determination of reducing power and ability to inhibit xanthine oxidase activity in vitro of determination method of Chrysanthemum indicum var.aromaticum stems antioxidant activity of different polar parts carries on the comprehensive evaluation results show that ethyl acetate parts of oxidation resistance,the strongest chloroform part strongest inhibition of xanthine oxidase activity...
