Objective:To test the function of Zhitongxiaojietang in the hyperplasia of mammary gland tissue of rats,to verify the curative effect of Zhitong-xiaojietang,bying observing the changes of pathologyicl morphology of r-at with hyperplasia of mammary gland,sex hormone receptors expression o-frat with hyperplasia of mammary glands.And then discuss the functional mechanism of Zhitongxiaojietang curing mammary gland hyperplasia from t-heperspective on adjustment of sex hormone receptors expression.Methods:Select 70 SPF-grade female SD rats and divide them at random int-o control group,model confirmed group,Zhitongxiaojietang groups of larg-e,medium,and low doses,as well as Rupixiao positive control group.Then established animal model of mammary glands with the combination of estra-diolbenzoate and progesterone.After the success of the building,respect ivelyfor drug treatment,Zhitongxiaojietang groups according to the conce-ntration of large,medium and low dose lavage,positive control group la-vage mixed suspension.The normal group and model group was given amount of distilled water to fill the stomach,for 30 consecutive days.Fasting c-an not help water 12 hours after the last treatment,by intraperitoneal injection of 10% chloral hydrate anesthesia for rats,and take the secon-d of right on full breast,observate pathological morphological changes of the breast tissue under optical microscope.Immunohistochemical staini-ng was used to observe the expression of estrogen receptor(FSH),progest erone receptor(T)in rat mammary gland.At the same time,the content of th-e receptor was semiquantitative analyzed.Results:1.Zhitongxiaojietang impact on the general condition of rats:com-pared with normal group rats,the model group there was a significantly increase in the nipple height and diameter of the second pairs of breast-s(P<0.01).After giving a corresponding drug gavage treatment,each grou-p rats nipple height and diameter than the model group significantly reduced(P<0.01).With Zhitongxiaojietang group of low dose is particular-ly significant.Compared with Rupixiao group,the Zhitongxiaojietang larg-e dose group had no significant difference.2.The effect of Zhitongxiaoj-ietang on pathological morpholog ical of rats:Normal group:Ductale pith-elium of breast arrayedregularly.No expansion was in ductal cavity and acinar cavity.The number of lobule of mammary gland and the gland alveol-usin lobule of mammary gland were very few.Model group:Hyperplasia of mammary glands is diffuse.Hyperplasia of the Gland alveolus and ductus was in great number.Gland alveolus in lobule o-f mammary gland was intensive and inequality of size.Obviously,there wa-s expansion in ductal cavity and acinar cavity.And so much secretion wa-s in intracavity.After treatment with drug lavage,low dose group of Zhi-tongxiaojietang does not change significantly,middle and high dose gro-up compared with model group,the number of the Gland alveolus was few a-ndthe secretion was so little.Hyperplasia of the Gland alveolus in Rupi-xiao control group was in different degree.However,expansion in ductal cavity and acinar cavity was not obvious.Rats compared with normal group,model group rats breast tissue in the FSH expression significantly highr and in the T expression significantly down(P<0.01).After drug gavage tr-eatment the pain subsides knot decoction high dose group can significan-tly reduce breast rights in breast tissue in rats of the expression of FSH and Tprotein(P<0.05 or P<0.01).And better than that of breast elim-ination group(P<0.05).Conclusion:Zhitongxiaojietang can reduce congestion,edema of the breast,improving mammary gland hyperplasia;Can adjust mammary glands FSH and T protein expression,then treatment of rat mammary gland hyperplasia.
