| Objective To explore the possible relationship between Recurrent Aphthous Ulcer(RAU) and the gene polymorphism of TGF-β1-509T/C site and IL-10-1082A/G site.Methods We collected 2ml peripheral venous blood from 138 patients with recurrent oral ulcer and 124 normal controls, and then extracted the DNA tissue from bloodstream. All the subjects’ TGF-β1-509T/C site and IL-10-1082A/G site have been detected by RFLP-PCR and SSP-PCR. The Hardy-Weinberg equilibrium can inspect groups’ representative. Chi-square test used to calculate each site of genotype frequency and allele frequency, with P<0. 05 for the difference is statistically significant.And then we use odds ratio(OR) 95% confidence interval(95% CI) to estimate relative risk.Results 1.In the control group, the genotype frequencies of TGF-β1-509T/C site and IL-10-1082A/G site conform with the Hardy-Weinberg equilibrium, and have a group’s representative. 2. According to the results of TGF-β1-509T/C site’s genotype frequency,compared with the disease group and the control group, CC, CT, TT genotype frequency is χ2=7.273, P< 0.026;and C, T allele frequency is χ2= 4.671, P<0.031.Compared with CC genotype, the OR value of genotype CT= 1.231(95% CI=0.702 ~ 0.702), and the OR value of the genotype TT = 2.482(95%CI=1.250 ~1.250). Used C allele as a standard, the OR value of T allele = 1.465(95%CI=1.036~1.036). 3.According to the results of IL-10-1082 site’s genotype frequency,compared with the disease group and the control group,AA,AG,GG genotype frequency is χ2=14.569, P<0.001;and A,G allele frequency is χ2=16.387, P<0.000.Compared with AA genotype, the OR value of genotype AG=1.391(95%CI=0.808~2.396),and the OR value of the genotype GG=4.165(95%CI=1.944~8.924). Used A allele as a standard, the OR value of G allele=2.134(95%CI=1.474~3.089).Conclusion TGF-β1-509T/C site and IL-10-1082A/G site are associated with the risk of RAU. The recurrent oral ulcer risk of the T allele which with TGF-β1-509T/C site is 1.465 times as higher as those with the C allele.The recurrent oral ulcer risk of the G allele which with IL-10-1082A/G site is 2.134 times as higher as those with the A allele. |
