BACKGROUND AND OBJECTIVE: Recurrent aphthous stomatitis (RAU) is acommon condition characterised by recurrent episodes of oral ulceration. Three clinicalforms are described; minor(MiRAU), major (MaRAU) and herpetiform ulceration(HU).MiRAU accounts for80%of RAUpatients. The ulcers are usually less than5mm indiameter, occur on the non-keratinised mucosa and heal spontaneously,without scarring, in7–14days. Some reports in the literature indicate that RAU may have immunological,psychological, genetic and microbiological bases。Genetic factors play an important role inthe development of RAU. More than forty percent of patients report a positive familyhistory and parental RAU status strongly influences the probability of an individualsuffering RAU.Furthermore, there is significantly higher disease concordance inmonozygotic than dizygotic twins.Objective:The purpose of the present study was to investigate, using Fisher exact testor chi-square test, a possible association between the functional IL-1B+3954(C/T) andIL-1B-511(C/T)genetic polymorphism and MiRAS in a sample of patients。Design:thirty-two consecutive subjects affected by minor forms of RAU and32healthy volunteers were genotyped. To investigate the association between the singlenucleotide polymorphisms and risk of RAU. P-values less than0.05were consideredsignificant.Materials and Methods:swabs were used for collection of cheek epithelial cells fromRAU patients and volunteers. One swab was used per cheek.Volunteers and patients wereinstructed to rub the inside of a cheek, swirling the swab fifteen times while so doing. Swabsamples were cut off to length and stored in15ml conical bottom tubes until extracted(overnight) at room temperature. DNA were extracted using Swab Gen DNA kit. DNAsamples were evaluated by measurement of optical density at260nm (OD260) and at280nm (OD280). To test the genotyping of DNA we choose PCR and gene sequencing.Conclusion: Our findings demonstrate that polymorphisms of IL-1B-511wereassociated with an increased risk of RAS development; the distribution of IL-1B+3954in thecontrol group was not statistically different from MiRAU group.Our findings also give additional support to a genetic basis for RAU pathogenesis.
