| Purpose: This study focuses on the Tang Yi Sheng Type from theories, clinical features ?and experiment. Specifically, the clinical observation to light T2 DM, the TUNEL apoptosis detection of ?-islet cells for Wistar rats with T2 DM, and the effect on expression of PDX-1 in the pancreases.Approaches: Clinical research: There are 60 patients with light T2 DM who have heat accumulating inside, and deficiency of spleen and kidney. To classify them into two groups, one is experimental group(30 patients using Tang Yi Sheng Type ?) and another one is control group(30 patients using metformin). Subsequently, to do comparison with pre-treatment for each group, in terms of clinical symptoms and signs, glucose and lipid metabolism, body mass index(BMI index), Hb A1 C, insulin and C-peptide level, insulin resistance(IR), and ?-cell function index.Experimental research: In order to establish T2 DM rats model, inject STZ into Wistar rats fed by high-sugar and high-lipid. After that, the study observes the influence of Tang Yi Sheng Type ?on these Wistar rats from several aspects: glucose and lipid metabolism, FINS, IR and BMI index, the TUNEL apoptosis detection level of ?-islet cells, and expression of PDX-1 in the pancreases based on Western Blot.Results: Tang Yi Sheng Type decreases the glucose and lipid metabolism level and the ?BMI index of patients with light T2 DM. Additionally, it benefits the ?-cell function index in basal condition, and improves the insulin sensitivity. Moreover, it also reduces the apoptosis detection level of ?-islet cells, and enhances the expression of PDX-1 in the pancreases.Conclusion: Tang Yi Sheng Type ?is beneficial to patients with light T2 DM. To be specific, it has higher safety, and reduces IR. The theory of Tang Yi Sheng Type might ?be relevant to the reduction of apoptosis detection level of ?-islet cells, and the improvement of the expression of PDX-1 in the pancreases, which is worthy of further research. |
