Objective: This study is based on the project, " Study on the Antioxidation Mechanism of Idiopathic Pulmonary Fibrosis by Multiple Ways of Expectorant Herbs", funded by the national natural science fund. According to the requirements of the research and teamwork, fritillaria cirrhosa was chosen as the research subject, based on Bleomycin-induced Pulmonary fibrosis of rats, to observe representation of rat pulmonary tissue in the aspects of pathology, oxidative stress and inflammatory reaction in the sole intervention of fritillaria cirrhosa and analyze its mechanism, in order to provide more evidences for clinical medication and the research about the mechanism of IPF.Method: Sixty male healthy Wistar rats in a SPF grade were selected and fed for 3 days for adaptation, then random number table method was applied to classify these rats into several groups: high-concentration fritillaria cirrhosa group(Group A), middle-concentration fritillaria cirrhosa group(Group B), low-concentration fritillaria cirrhosa group(Group C), Pirfenidone group(Group D), model control group(Group E) and blank control group(Group F). Under anesthesia of 10% chloral hydrate, bleomycin was dropped into the windpipe of rats from Group A to Group E based on 5mg/kg to establish rat fibrosis model, and normal saline in same volume was dropped into the windpipe of rats from Group F under the same condition of anesthesia. Rats were given expected drugs by gavage according to their weights every day since the fourth day after modeling. They were killed after 28 days' medicine and pulmonary tissue was taken to do pathology observation under the light microscope; content of GSH, MDA AND SOD in the issue was detected with BCA method; content of TNF-?, TNF-?1, Nrf2, PPARs and VEGF in the issue was measured with ELISA. For experimental data, SPSS 17.0 was applied to conduct statistical treatment.Result: pathological section showed that, except the blank control group, each group showed different degrees of interstitial lung disease pathological changes and the model control group was the worst. All the treatment groups performed better than the model control group. fritillaria cirrhosa showed its therapeutic effects in the testing of many items of rat lung tissue, and Fritillaria cirrhosa even had better effect than pirfenidone in some items. Different concentrations of fritillaria cirrhosa varied in effect and middle or high concentration performed better in most items.Conclusion: Fritillaria cirrhosa can intervene Bleomycin-induced pulmonary fibrosis of rat through a multi-channel and its effects may be related with oxidative stress, inflammatory reaction, vascular proliferation,etc., which provides more evidences for clinical medication and the research about the mechanism of IPF.
