Fritillaria cirrhosa D.Don is one of Fritillaria L, that can inhibits cough significantly. It was reported that Fritimine inhibits cough, It has Effects of Immunity,Antitusis,Expectoration and Antiasthma. There are three parts in this paper. First, the Fritimine has been extracted using biochemistry technology from Fritillaria cirrhosa D.Don. Second, experiments of antitusis and antiasthma of Fritimine have been carried out in animals. Last, by testing the content of second messenger cAMP and NO in trachea tissue, the mechanism of actions above-entioned has been discussed on mice.In the research, Fritimine has been extracted from Fritillaria cirrhosa D.Don by biochemistry technology of alkaline decomposition. acidification. extraction and confirmed, choose NH3.H2O and SO2 make mice cough. Then, NO content in serum and trachea was determinated with the method of spectroprotometry and cAMP content trachea by Competitive protein binding assay method, by testing the content of second messenger cAMP and NO intrachea tissue,the might mechanism of Fritimine of actions above-entioned has been discussed.The results of antitusis and antiasthma experiments show Fritimine can significantly control the cough of mice caused by NH3.H2O and SO2 and has strong resolving phlegm and antiasthmatic functions when it is given in 0.054g/kg and 0.0054g/kg doses and by oral administration. It show Fritimine can enhance the respiratory function in high dose and low dose and inhibits significantly the specifc change in serum and trachea of mice when it is given in high dose on male mice. Competitive protein binding assay indicates the level of cAMP is improved when Fritimine is given by oral administration. Then, NO content in serum and trachea was determinated with the method of spectroprotometry. The result of the test shows that Fritimine can significantly reduce the quantity of NO in serum and trachea, in a word , Fritimine is one of active principles of Fritillaria cirrhosa D.Don. |
