Objective:By detecting the polymorphism of AGT 7 and AT1 R 2 loci in patients with essential hypertension between different TCM syndrome types, to discuss the distribution of genotypes and allele frequency between the different syndromes EH group and the control group of 9 loci.And to explore the effect of the interaction to EH patients with different TCM syndrome types of these loci.Methods:According to TCM syndrome types, which are: Liver-fire hyperactivity type,Yin deficiency with yang hyperactivity type,Phlegm-damp retention type,Dual deficiency of yin and yang type,we select one hundred patients with essential hypertension in each type.,and another one hundred normal persons were selected for the control group. The Improved Multiple Ligase Detection Reaction(im LDR) is used to detect the polymorphisms of AGT gene(rs5049,rs2148582,rs4762,rs5051,rs699,rs11568020,rs5050) and AT1 R gene(rs5186,rs5182). We use the method of Multifactor Dimensionality Reduction(MDR) to analyze the effect of AGT and AT1 R gene interaction on EH patients with different TCM syndrome types.Results:1. Comparion with EH group and the control group for frequency of gene polymorphism of AGT and AT1 R gene 9 loci,there are significant differences in the frequency of gene polymorphism of the AGT gene rs5049,rs5051,rs2148582,rs699, r rs4762(p<0.05). C and T allele of AT1 R gene rs5182 loci have significant difference between EH group and the control group(p<0.05).2.Compared with different syndromes EH group and the control group for genotypes and allele frequency of AGT and AT1 R gene, there are significant differences in the polymorphism of the AGT gene rs5049 in phlegm-damp retention type and yin deficiency with yang hyperactivity type(p<0.05). There are significant differences in the polymorphism of the AGT gene rs2148582, rs5051 and rs699 in phlegm-damp retention type and dual deficiency of yin and yang type(p<0.05). There are significant differences in the polymorphism of the AGT gene rs4762 and rs5050 in yin deficiency with yang hyperactivity type(p<0.05). There are significant differences in the polymorphism of the AGT gene rs11568020 in liver-fire hyperactivity type(p<0.05).AT1 R gene rs5186 polymorphism have no significant difference(p >0.05).There are significant differences in the polymorphism of the AT1 R gene rs5182 in dual deficiency of yin and yang type(p<0.05).3. By apply MDR,the best diagnostic model of EH group, liver fire hyperactivity,phlegm-damp retention, yin deficiency with yang hyperactivity, and dual deficiency of yin and yang are the interaction of AGT rs5051 and AT1 R rs5182 genes, the interaction of AGT rs5049,AGT rs2148582 and AT1 R rs5182 genes, the interaction of AGT rs5050, and the interaction of AGT rs4762, AGT rs5050 and AT1 R rs5182 genes,respectively, all with statistical significance p <0.05.Conclusion:1. AGT gene rs5049,rs5051,rs2148582,rs699 and rs4762,these 5 loci may associated with the pathogenesis of EH. AT1 R rs5182 gene T allele may associated with the pathogenesis of EH.2. To research AGT and AT1 R gene 9 loci, we found that in addition to the AT1 R gene rs5186 loci, the rest loci are related to EH different syndrome types in different degree.Especially, the AGT gene rs5049, rs2148582, rs699 and rs5051 these four loci were significantly associated with EH phlegm-damp retention type.3. There is a obvious interaction between AT1 R gene rs5182 and AGT different loci in liver-fire hyperactivity type,Phlegm-damp retention type and dual deficiency of yin and yang type.AGT gene rs5050 loci is the susceptible factors for EH of yin deficiency with yang hyperactivity type in Hefei Han.The interaction between AGT and AT1 R gene polymorphisms serves as the cause in attacks of EH patients with different TCM syndrome types.
