Objective: Through the nm23 H1 and integrin alpha 3,beta 1 expression and correlation study in endometriosis,we expect to have a better understanding with nm23-H1 gene and cell adhesion pathways related molecular integrin alpha 3,beta 1and the relationship between endometriosis.It is hope to provide new train of thought and basis to the clinical diagnosis,prognosis and treatment of endometriosis.Methods: Collect 47 cases of normal endometrium intrauterine membrane specimens and 42 cases of endometriosis specimens,all specimens in triplicate.One for general Trizol one-step extraction of total RNA,reverse transcription to total c DNA,using real-time fluorescent quantitative RT-PCR technology to detect nm23-H1 and integrin alpha3,beta 1 gene m RNA expression level;Another used to extract total protein in the organization,using Western Blot technique to detect nm23 H1 and integrin alpha 3,beta 1 gene protein expression level;The last used for laser capture microdissection technology,capture endometrial epithelial cells.trace of RNA extraction kit is utilized to extract total RNA in the cell,and then uses the real-time fluorescence quantitative RT-PCR technique to detect nm23 H1 and integrin alpha 3,beta 1 m RNA expression levels of genes in endometrial epithelial cells.Results: 1.Regular qRT-PCR to detect gene expression:(1)The endometriosis group and control group in nm23-H1 gene m RNA expression levels were0.42±0.19,0.96±0.98.The endometriosis groups nm23-H1 gene m RNA expression level is lower than the normal control group,the differences between the two groups was statistically significant(t = 3.738,P = 0.000,P < 0.05).(2)The endometriosisgroup and the control group of integrin alpha 3 gene m RNA expression levels were6.67±1.28,6.46±2.25.Compared the two,the differences between the two groups was no statistically significant(t = 0.53,P = 0.59,P > 0.05).(3)The endometriosis group and the control group of integrin beta 1 gene m RNA expression levels were10.59±2.72,8.39±2.81.The endometriosis groups of integrin beta 1 gene m RNA expression levels higher than normal control group,the differences between the two groups was statistically significant(t = 3.75,P = 0.000,P < 0.05).2.Western Blot technique to detect protein expression:(1)The endometriosis group and control group in nm23-H1 gene protein expression levels were0.71±0.41,1.06±0.73.The endometriosis group nm23-H1 protein expression levels lower than normal control group,the differences between the two groups was statistically significant(t = 2.557,P = 0.013,P < 0.05).(2)The endometriosis group and the control group of integrin alpha 3 gene protein expression levels were0.50±0.29,0.60±0.33.Compared the two,the differences between the two groups was no statistically significant(t = 1.43,P = 0.16,P > 0.05).(3)The endometriosis group and the control group of integrin beta 1 gene protein expression levels were0.98±0.45,0.77±0.33.The endometriosis groups of integrin beta 1 gene protein expression levels compared with normal control group,the differences between the two groups was statistically significant(t = 2.36,P = 0.021,P < 0.05).3.The laser capture microdissection technology to detect the expression of target genes in endometrial epithelial cells:(1)The endometriosis group and control group in endometrial epithelial cells nm23-H1 gene m RNA expression levels were0.46±0.38,1.37±0.95.In the endometriosis groups of endometrial glandular epithelial cells of nm23-H1 gene m RNA expression levels were lower than that of normal control group,the differences between the two groups was statistically significant(t= 3.597,P = 0.002,P < 0.05).(2)The endometriosis group and control group in endometrial epithelial cells of integrin alpha 3 gene m RNA expression levels were4.91±2.55,5.93±1.63.Compared the two,the differences between the two groups was no statistically significant(t = 1.44,P = 0.16,P > 0.05).(3)The endometriosis group and control group in endometrial epithelial cells of integrin beta 1 gene m RNA expression levels were 5.96±1.94,3.77±3.06.The endometriosis group of endometrial glandular epithelial cells of integrin beta 1 gene m RNA expression levels were higher than in normal control group.The differences between the two groups was statistically significant(t = 2.47,P = 0.02,P < 0.05).Conclusion: nm23-H1 as a tumor metastasis suppressor gene expression in endometriosis,prompt the process involved in the occurrence and development of endometriosis.nm23-H1 can be based on the related signaling pathways of cell adhesion,regulating adhesion molecules,such as calcium adhesive element,? integrin,strengthen the adhesive force between cells,inhibit tumor cell metastasis.In endometriosis,integrin beta 1 gene expression level of change,suggest there is a certain correlation with the expression of gene nm23-H1.In a word,further explore the nm23-H1 gene in endometriosis,help in the early diagnosis,treatment and prognostic evaluation of the diseases.
