Objective:To explore the effect of different concentrations of TNF-alpha and TNF-alpha antibody on the expression of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclasts.Methods:Mice RAW264.7 cells were induced differentiation into osteoclast in vitro.And the osteoclastogenesis was detected by tartrate resistant acid phosphatase staining.Then the osteoclasts were divided into three groups.The control group,the TNF-alpha intervention group(treated with different concentrations of TNF-alpha for 48h)and the TNF-alpha antibody intervention group(treated with different concentrations of TNF-alpha antibody for 48h).The mRNA and the protein expression levels of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast were respectively examined by real-time PCR and Western blot.Results:Mice RAW264.7 cells were induced by RANKL for six days,then TRAP staining proved the formation of multinucleated osteoclasts.The mRNA expression levels of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast in TNF-alpha treatment group were significantly higher than these in the control group(P<0.001).The mRNA expression levels of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast in TNF-alpha antibody treatment group were significantly lower than these in the control group(P<0.001).The protein expression levels of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast in TNF-alpha treatment group were significantly higher than these in the control group(P<0.05).The protein expression levels of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast in TNF-alpha antibody treatment group were significantly lower than these in the control group(P<0.05).Conclusion:TNF-alpha can improve the expressions of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast;TNF-alpha antibody can reduce the expressions of integrin alpha-v,integrin beta-3 and V-ATPase on osteoclast.These suggested that TNF-alpha can improve osteoclastic bone absorption by increasing the expressions of integrin alpha-v,integrin beta-3 and V-ATPase.
