Objective: By comparing the mRNA expression level of nm23-H1 and p53 gene in endometriosis and in normal control,we investigate the roles of nm23-H1 and p53 gene in the pathogenesis and development of endometriosis,analyze the correlation between expression of the two genes,In order to further study its role and mechanism in endometriosis.Methods: We detected the differential expression of nm23-H1 gene and p53 gene,which existed in endometriosis specimens and normal endometrial tissue specimens,using real-time quantitative PCR method.And analyzed the correlation between expression levels of the two genes.The method of Western Blot was taken to detect the expression of nm23-H1 gene and p53 gene.Results: 1.Regular qRT-PCR to detect gene expression:The endometriosis group and control group in nm23-H1 gene mRNA expression levels were0.42±0.19,0.96±0.98;the expression levels of p53 mRNA in 41 endometriosis groups and in 47 control groups were 0.44 ± 0.5 and 0.86 ± 1.05,respectively;the mRNA expression levels of nm23-H1 and p53 gene were lower than those of normal control group,The differences between the two groups was statistically significan(t=3.738,P=0.000;t=2.405,P= 0.019).2.The laser capture microdissection technology to detect the expression of target genes in endometrial epithelial cells: The19 cases of endometriosis group and 16 cases of control group in endometrial glandular epithelial cells captured by Laser capture microdissection,the nm23-H1 gene mRNA expression levels were 0.46±0.38,1.37±0.95,respectively;p53 gene mRNA expression levels were 0.55±0.43 and 1.39±1.36 in endometriosis group and control group,respectively;endometrial glandular epithelial cells of endometriosis of nm23-H1 gene and p53 gene mRNA expression levels were lower than those of the normal control group,the difference between the two groups has statistical significance(t = 3.597,P = 0.002;t=2.349,P = 0.031).3.Western Blot technique to detect protein expression: The expression level of nm23-H1 protein was 0.71±0.41 in 39 endometriosis groups,and 1.06±0.73 in 35 control groups;the expression levels of p53 protein in 39 endometriosis groups and in 35 control groups were 0.65±0.38 and 0.96±0.39,respectively;the protein expression levels of nm23-H1 and p53 gene were lower than those of normal control group,and there were statistically significant differences between the two groups(t=2.557,P=0.013;t=3.483,P=0.001).4.The relevance of nm23-H1 genes and the p53 gene expression: Both methods detect the mRNA expression level of nm23-H1 gene and p53 gene in endometriosis group have a certain correlation(r = 0.356,p = 0.022;r = 0.604,p = 0.013),two gene expression in the control group have no correlation(r = 0.165,p = 0.268;r = 0.188,p = 0.486).There was certain correlation between the protein expression levels of nm23-H1 gene and p53 gene in endometriosis group(r=0.571,P=0.000).in normal control group,There were no correlation between the protein expression levels of the two gene(r=0.257,p=0.142).Conclusion: The decline of mRNA and protein expression levels of nm23-H1 gene and p53 gene may be associated with the occurrence of endometriosis,and nm23-H1 gene may play a role in endometriosis by the way of p53 partly,the exact mechanism is unclear.it's remains to be further studied.
