| Purpose:To study the influence of electro-acupuncturing with selecting acupoint along meridians on cardiac chloride channel CLC-3, CLC-4 in rats with myocardial ischemia induced by isoprenaline hydrochloride(ISO). And to explore the mechanism of electro-acupuncturing with selecting acupoint along meridians, in order to provide scientific experimental basis for clinical acupuncture treatment of myocardial ischemia.Material and method:50 male SPF rats were randomly divided into five groups with a random number table, i.e., the blank group, the model group, the Neiguan(PC6) group, Lieque(LU7) group, non-acupoint(the midpoint of Shenque(RN8) and Tianshu(ST25)) group, 10 in each group. ISO was subcutaneously injected in all groups except the blank group to establish the myocardial ischemia model. Rat in blank group were subcutaneously injected with an equal volume of normal saline. After successful modeling,EA at each acupuncture point was applied for rats in the Neiguan group, the Lieque(LU7) group and the non-acupoint group energized needle retention 15 min once a day for 7 successive days, while the model group was just fixed without EA. Finally rats were sacrificed after 30 min of EA on the seventh day. The sampling was taken. The indices such as superoxide dismutase(SOD) activities, lactate dehydrogenase(LDH), morphology changes of apoptosis cells and the change of cardiac chloride channel CLC-3 and CLC-4 protein expression were observed, respectively.Results: 1. Compared with the blank group, LDH contents of the modeling group,the Lie Que group and the Non-acupoint group are significantly increased(P<0.01), compared with the model group, the serum LDH contents were significantly reduced(P<0.01). 2. Compared with the blank group, SOD contents of the modeling group,the Lie Que group and the Non-acupoint group are significantly reduced(P<0.01), compared with the model group, the serum SOD contents were significantly increased(P<0.01). 3. Compared with the blank group, the level of apoptotic cells of the modeling group,the Lie Que group and the Non-acupoint group are significantly increaed, but the level of apoptotic cells in the Neiguan group is not obvious. 4. Compared with blank group, there is a significant increase in the contents of CLC-3ã€CIC-4 CL- channels protein in the modeling group,the Lieque group and the Nonacupoint group(P<0.01).And there is no significant differences were found between the blank group and the Neiguan group( P>0.05).Conclusion: 1. Acupuncture at Neiguan(PC-6) can inhibit the apoptosis of the myocardial cells in experimental myocardial ischemia of rats. 2. It effectively develops the rats myocardial ischemia and reduce LDH contents and increase SOD contents in serum by acupuncture at Neiguan acupuncture point. 3. It is one of possible mechanisms to reduce myocardial harm of ischemia, with regulating the expression of myocardial tissue chloride channel CLC-3 and CIC-4 protein. 4. The effect of acupuncture point tired specificity is exist. |
