Purpose: The study to observe the electro-acupuncture stimulate the heart meridians simultaneously to the effect of electrocardiograph,cardiac ischemic area,serum creatine kinase and norepinephrine,dopamine,serotonin in acute myocardial ischemia(AMI)rat,in order to Investigate the effect material basis of hippocampus involved in electroacupuncture in the treatment of myocardial ischemia.Methods: According to random number table method,the SD rats were randomly divided into sham group,model group,acupuncture group,6 rats in each group.Model group and acupuncture group were established,which is cut off the left anterior descending coronary artery ligation.The acupuncture group the heart meridian "Shen Men(HT-7)-Tong Li(HT-5)",were selected and electroacupuncture(EA)with 1m A intensity,2Hz disperse-dense wave,for 30 min a day in 3 days.Powerlab16 is used to record the heart rate(HR),ECG,ECG-ST,T wave.After acupuncture,collecting dislysate of hippocampal CA1 region and detecting the content of norepinephrine,dopamine,serotonin.Enzyme-linked immunosorbent assay(ELISA)is used to detect serum creatine kinase content in peripheral blood plasma.The cardiac ischemic area of rats is stained by 2,3,5-triphenyltetrazolium chloride(TTC).We analyzed the differences of HR,ECG,serum creatine and monoamine neurotransmitters in each groups;the correlation between norepinephrine,dopamine,serotonin,serum creatine kinase in rats.Results:(1)Compared with the sham group,the HR?ECG,ECG-ST,T wave were significantly increased in the model group(P<0.01);Compared with the model group,the HR?ECG,ECG-ST,T wave were significantly decreased in the acupuncture group(P<0.01).(2)Compared with the sham group,the cardiac ischemic area weresignificantly increased(P<0.01);Compared with the model group,the cardiac ischemic area were significantly decreased in the acupuncture group(P<0.01).(3)Compared with the sham group,the serum creatine kinase were significantly increased(P<0.01);Compared with the model group,the serum creatine kinase were significantly decreased in the acupuncture group(P<0.05).(4)Compared with the sham group,the norepinephrine were significantly increased(P<0.01),dopamine,serotonin were significantly reduced(P<0.01);Compared with the model group,the norepinephrine were significantly decreased(P<0.01),dopamine,serotonin were significantly increased(P<0.01).(5)Pearson correlation analysis showed that the norepinephrine content in the rat hippocampal CA1 region was positively correlation with serum creatine kinase(r= 0.587,P<0.01);the dopamine and serotonin were negatively correlation with serum creatine kinase(dopamine,r=-0.597,P<0.01;serotonin,r=-0.7602,P<0.01).aConclusion: 1.Electro-acupuncture significantly improve the ECG and reduce ischemic area,serum creatine kinase of acute myocardial ischemia rats,The results suggesting that electro-acupuncture at the heart meridian has the effect of improving acute myocardial ischemia.2.Monoamine neurotransmitters are the effectors that the hippocampus involved in the effect of the electro-acupuncture intervene the acute myocardial ischemia.
