Study On The Method Of Improving The Ratio Of Clamp Connection Of Aurieuzaria Auricular

Objective: To obtain a better method of improving Aurieuzaria auricular clamp connection hyphae number,the five species “H15”,“H29”,“XK1”,“XK10”,“SL15” of Aurieuzaria auricularas were choosed for researching higher products and better breeds under the condition of different nutrients and environments.Methods:(1)The effects of different nutritional conditions on the growth of Auricularia auricula.Take the five species strains of Aurieuzaria auricular into the plate centre with different carbon and nitrogen sources,and amino acids(inserting cover slip)respectively.The deal with basic medium that serving as the control groups were cultured in the temperature of 25 ?.(2)The effects of different light qualities and temperatures on the clamp connection mycelium growth of Aurieuzaria auricular.Take the five species strains of Aurieuzaria auricular into the plate centre of PDA medium(inserting cover slip).The dealing group were taken into cultivating incubator with LED light,meanwhile the control groups were treated in dark environment for 24 h.In turn,the PDA medium(inserting cover slip)were taken to the place with 30 cm distance to the 30 W UV light source to be radiated for 1 min,2 min,3 min,4 min,5 min,respectively.The samples were protect by isolating light after radiation so that avoid from photoreactivation,the groups no radiation of UV serve as control.All the deals were cultured in the temperature of 25 ?.The deals were repeat each three days.Take the five species strains of Aurieuzaria auricular into the plate centre of PDA medium(inserting cover slip)and treat with the temperatures of 20 ?,25 ?,30 ?,35 ?,respectively.(3)The optimization of the medium.According to the previous experimental results,select the “H29” as the varieties of Aurieuzaria auricular for further study,and to determine the carbon source,nitrogen source and amino acid,that have a obvious role on clamp connection,were sucrose,beef extract,alanine respectively.First the single factor experiments were taken to determine the best concentration,and then the orthogonal experiments were taken again.All the tests take six repeats,cultivating 12 d,and then take out the inserting cover slips to observe in the microscope.Results:(1)To compare to glucose(monosaccharide)as carbon source,when “H29” using sucrose as carbon source,the clamp mycelial growth of about 2/3 ratio;“H15” with soluble starch as carbon source growth more than 1 times;“XK1” with maltose as the carbon source growth of nearly 2 times;five varieties of Aurieuzaria auricular in beef extract as nitrogen source,clamp connection hyphae(P<0.05)rate was significantly higher than that other groups,especially “H15” and “H29” are the most significant(P<0.05).The five species clamp connection hyphae of Aurieuzaria auricular have improved significantly in the culture of adding alanine glutamic acid into medium.(2)The red,blue and purple wave length radiation on the formation of clamp connection hypha to “H29” and “SL15” have increased significantly(P<0.05).However,for the“H15”,“H29”,“XK1”,“SL15”,the formation of clamp connection hyphae were increased in the radiation of UV in 1 min,4 min,5 min,3 min respectively.For the species “H15”,“H29”,“XK1”,“XK10” of Aurieuzaria auricular,the ratio of clamp connection hyphae were the highest than that others in the temperature of 25 ?(P<0.05).But the “H15” was opposite.(3)The appropriate concentrations of sucrose,beef extract,alanine could improve the “H29” clamp connection hyphae ratio,and the optimal combination is 20 g/L sucrose,6 g/L beef extract,3.5 g/L alanine,rspectively.Conclusion: Adding appropriate concentrations of carbon sources,nitrogen sources and amino acids in the culture medium can improve the ratio of the Auricularia auricula clamp connection hyphae.The effects showed as follows: carbon sources?nitrogen sources ?amino acids.The effects of blue and red wave lights on clamp connection hyphae are better than other wave lights.UV radiation time should be controlled in about 3 min,it will greatly reduce the survival rate of strains.Temperature should be controlled at 25 ?.lower or higher temperature are not conducive to the growth of mycelium.The results of orthogonal experiments showed that the addition of sucrose 20 g/L,beef extract 6 g/L,alanine 3.5 g/L,in the basic culture medium,could significantly increase the ratio of clamp connection to “H29” species.
