Acute renal failure(ARF)is a group of clinical syndromes characterized by rapid decline in glomerular filtration rate, accompanied by the accumulation of nitrogen waste, serum creatinine(Cr)and blood ure nitrogen(BUN)retention, disturbance of water, electrolyte and acid-base balance, and systemic complications. This disease is common in small animal clinical, and the serious ARF can lead to animal death. In this paper the animal model of canine ARF was established by injection of medicine to dogs, and two methods were used for treatment of these dogs to evaluate the effect of the methods, which can provide come guidance for treatment of canine ARF.The following results have been achieved in this study.1.Establishment of acute renal failure model in dogs In this text, eight healthy mixed dogs were chosen as experimental animals(about 5 kg per dog).Gentamicin was administrated continuously by subcutaneous injection with the dose of 40 mg/kg·d to 60 mg/kg·d(40 mg/kg·d at first, after 5 days the dose changed to 60 mg/kg every 12 h).After administration of the medicine foe 5 to 11 day,the animals showed vomiting, oliguria and other clinical symptoms. The hematological indexes such as blood urea nitrogen(BUN),creatinine(Cr),neutrophil gelatinaseassociated lipocalin(NGAL)and cyctatin C(Cys C)etc.were significantly increased. The histopathological observation of the kidney showed obvious vacuolar degeneration in the kidney tubules and atrophy of the renal glomerulus. These results suggested that kidney units of dogs had parenchymal damaged and the kidney function had reduced, and these explains that the acute renal failure model in dogs were established successfully.2.The treatment effect of two therapeutic methods for acute renal failure in dogs Eight dogs with ARF were randomly divided into intravenous infusion group and peritoneal dialysis group, each for 4 dogs, and these 2 group were treated by therapies of venous infusion and peritoneal dialysis.By comparing the two groups of dogs with clinical signs, concentration of the biochemical indexes(BUN,Cr,NGAL and Cys C atc.),and the pathological changes of kidney, the therapeutic effect of two treatment methods for dogs with ARF were evaluated. The results showed that, compared the dogs in intravenous infusion group, dogs in peritoneal dialysis group were recovered earlier in mental status and appetite, and the biochemical indexes were also declined earlier, and the degree of glomerular atrophy was lighter than that of dogs in intravenous infusion group. The results in this paper can provide a new thought for establishment of ARF model in dogs, and provide some valuable references for the clinical treatment of ARF in dogs by compared the therapeutic effect of two therapies. |
