This study was to explore Ala-Gln and MOS of weaned piglets stress production performance,intestinal morpHology structure,the influence of the intestinal barrier function and blood biochemical indexes,The results would provide a theoretical basis for the applications of Ala-Gln and MOS in the piglet feed.Experiment I: Effects of Alanyl-glutamine dipeptide and Mannan Oligosaccharides on growth performance and diarrhea rate in weaned pigletsA total of 162 piglets weaned at d 21 and with the similar genetic background were randomly allotted to 9 treatments with 3 replicates pertreatment and 6 piglets per replicate using a randomized complete-block design.The control group was subjected to receive the basal diet,and the other group was fed the basal diet supplementation 0.10%MOS,0.20%MOS,0.15% Ala-Gln,MOS+ 0.15% Ala-0.10% Gln,0.20%MOS+0.15%Ala-Gln,0.30%Ala-Gln,0.10%MOS+0.30% Ala-Gln,0.20% MOS+0.30% Ala–Gln.The experiment lasted 28 days.At the d 0,d 14 and d 28,piglets in every repeat were weighed and feed consμmption was also recorded for the computing of average daily feed intake,averaga daily gain,feed gain ratio and diarrhea rate.The results showed that:(1)d 21~35,basic diet adding 0.20% MOS and 0.20% Ala-Gln significantly reduced weaned piglets stress material weight ratio(P < 0.05),and d 35~49,basic diet adding 0.20% MOS and 0.20% Ala-Gln significantly improve piglets weaned stress on average daily gain(P< 0.05),and significantly reduce the weaned piglets stress material weight ratio(P< 0.05),basic diet adding 0.10% MOS and 0.10% Ala-Gln,0.20% MOS and Ala-0.20% Gln significantly improve piglets weaned stress average daily feed intake(P< 0.05),(2)No interaction appeared between Ala-Gln and MOS in improving growth performance when Ala-Gln and MOS were combined to use indiet of piglets.In conclusion,growth performance of piglets were improved by dietary MOS or Ala-Gln supplemen-tation and 0.20% MOS+0.30%Ala-Gln was an appropriate and feasible combination.Experiment II: Effects of Alanyl-glutamine dipeptide and Mannan Oligosaccharides on the intestinal morp Hology and the intestinal barrier function in weaned morpHology.Take double ligation on both ends of the cecμm middle insufflate samples,tinfoil,wrap with plastic wrap,together with the thread end,the other end of the stick label paper,liquid nitrogen preservation,used for determination of microorganisms.Take cecμm contents for microorganism determination,take the jejunμm mucosa for closely connected to the determination of the protein.Results showed:(1)d21~35,basic diet adding 0.10% MOS + 0.10% Ala-Gln significantly improved duodenμm villus height(P < 0.05),basic diet adding 0.10% MOS + 0.10% Ala-Gln significantly improve jejunμm villus height,and villus height/crypt depth(P<0.05),basic diet adding 0.20% MOS + 0.20% Ala-Gln significantly improve the ileμm villus height,reduce the depth of duodenal fossae,improved the duodenμm and ileμm villus height/crypt depth(P<0.05)(2)d 35~49,basic diet adding 0.10% MOS + 0.30% Ala-Gln significantly improved duodenμm villus is high,the lower the ileal crypt depth(P < 0.05),basic diet adding 0.20%MOS+ 0.20% Ala-Gln significantly improve jejunμm villus height,lower jejunμm villus height/crypt depth(P<0.05),basic diet adding 0.10%MOS+0.10%Ala-Gln significantly lower jejunμm crypt depth(P<0.05),basic diet adding 0.20%MOS + 0.20%Ala-Gln significantly improved the duodenμm and ileμm villus height/crypt depth(P<0.05).(3)d 35~49,the MOS and Ala-Gln exist interaction effect,mainly reflected in the duodenμm villus height,and villus height/crypt depth.(4)d 21~35,Experimental piglets jejunμm,neat,dense microvilli is longer,especially in the basic diet adding 0.10% MOS + 0.10% Ala-Gln,d 35~49,in the basic diet adding 0.20% MOS + 0.30% Ala-Gln,a longer jejunμm microvillus,neat,dense.(5)d 35 and d 49,closely connected protein group were significantly higher than that of control group.Above all,MOS and Ala-Gln of weaned piglets stress stimulates intestinal villi development,can improve the jejunμm microvillus structure,promote the jejunμm mucosa tight junction protein Occludin expression,and both have interaction effect on the intestinal villi.Experiment III: Effects of Alanyl-glutamine dipeptide and Mannan Oligosaccharides on blood biochemical index of weaned pigletsA total of 162 piglets weaned at d 21 of age and with the similar genetic background were randomly allotted to 9 treatments with 3 replicates pertreatment and 6 piglets per replicate using a randomized complete-block design.The control group was subjected to receive the basal diet,and the other group was fed the basal diet supplementation 0.10%MOS,0.20%MOS,0.15%Ala-Gln,0.10%MOS+0.15% Ala-Gln,0.20% MOS+0.15% Ala-Gln,0.30%Ala-Gln,0.10% MOS +0.30% Ala-Gln,0.20% MOS + 0.30% Ala-Gln.The experiment lasted 28 days.At the 14 th day and 28 st day,one piglets were vacuμm blood(8-10 ml)collection and separate serμm randomly selected from each repeating group.after the centrifuge separation of glutamate-puruvate transaminnase(ALT),glutamic-oxaloacetic transaminase(AST)et.Results showed:(1)d 35,basic diet adding 0.10% MOS + 0.10% Ala-Gln significantly improve piglets weaned stress of the vitality of SOD,GSH-PX(P<0.05),basic diet adding 0.20% MOS + 0.20% Ala-Gln significantly increase the content of weaned piglets serμm TP stress,the activity of ALT,AST,reduce the content of COR(P < 0.05),basic diet adding 0.10% MOS + 0.10% Ala-Gln significantly reduced the content of the COR in weaned piglets stress(P<0.05).(2)d 49,basic diet adding 0.20%MOS significantly improve the activity of serμm AST(P<0.05),basic diet adding 0.10% MOS+0.10%Ala-Gln significantly reduced the content of serμm COR(P< 0.05).
