| Salix is one of the excellent economic tree species in China.Salix has many advantages,such as rich resources,fast-growing,strong resistance,wide range of adaptation,beautiful shape and so on.Salix can not only produce timer and other forest products,beautify and improve the ecological environment,but also improve the ecological environment,greening and beautify the city landscape.It is the tree species that collect economic value,ecological value and ornamental value in one.Although most of Salix breeding through cuttings,there are still a few types of cutting difficult to breed.At the same time,branches of some Salix for mass production is limited,it is difficult to meet the production requirement.The plant micropropagation technology have the advantage of high productive coefficient,short cycle,fast breeding,it is of great theoretical significance to the biology research of Salix.And rooting seedlings transplanted is not limited by the season,so it is conducive to large-scale breeding.In recent years,the Salix in micropropagation technology has been studied,but its key technologies still need to be further optimized.So,in this study,we selected the Salix with high ornamental value and application value,such as Salix chaenomeloides ‘ Variegata ’ 、 Salix taraikensis 、 Salix matsudana ‘Chameleon’、Salix integra、Salix integra’Hakuro Nishiki’、Salix sinopurpurea、Salix psammophila 、 Salix argyracea,to carry out the optimization of the disinfection and sterilization method of explants,the screening of lateral buds inducing medium and rooting culture medium.And the best sterilization method was selected,the best basic medium for induction and rooting of lateral buds is decided,to provide technical support for the establishment of the aseptic system of Salix and the optimization of Salix micropropagation.1.The stem sterilization method of Salix chaenomeloides‘Variegata’:In this study,Salix chaenomeloides semi-lignified stem segments as experimental materials,by using 75% alcohol and different concentrations and different disinfection time of sodium hypochlorite for disinfection,disinfection analysis,combination of 75% ethanol 30s+15% sodium hypochlorite 20 min,Salix chaenomeloides germination rate was the highest,reached 100%,the pollution rate is relatively low,up to 10%,can be used as Salix the best explant disinfection method.2.Screening of medium for lateral buds of Salix,the results are:(1)Salix chaenomeloides best lateral bud induction medium is improved DKW,the germination rate is 87.50%;(2)Salix integra‘Hakuro Nishiki’ best lateral bud induction medium is 1/2WPM,the germination rate is 100%;(3)Salix taraikensis best lateral bud induction medium is improved DKW,the germination rate is 93.75%;(4)The Salix matsudana ‘Chameleon’ best lateral bud induction medium is 1/2MS,the germination rate is 94.44%;(5)Salix integra best lateral bud induction medium is improved DKW,the germination rate is 66.67%;(6)Salix purpurea best lateral bud induction medium is improved DKW,the germination rate is 100%;(7)Salix psammophila lateral bud induction medium is 1/2WPM,the germination rate is 100%;(8)Salix argyracea lateral bud induction medium is improved DKW,the germination rate is 100%.3.Screening the rooting medium of aseptic buds of Salix the results are:(1)Salix chaenomeloides best rooting medium is 1/2WPM,the rooting index is 72.91;(2)Salix integra‘Hakuro Nishiki’ best rooting medium is 1/2WPM,the rooting index is 244.28;(3)Salix taraikensis best rooting medium is improved DKW,the rooting index is 79.65;(4)The Salix matsudana ‘Chameleon’ best rooting medium is 1/2WPM,the rooting index is 130.27;(5)Salix integra best rooting medium is 1/2WPM,the rooting index is 183.12;(6)Salix purpurea best rooting medium is 1/2WPM,the rooting index is 136.51;(7)Salix psammophila best rooting medium is 1/2WPM,the rooting index is 165.31;(8)Salix argyracea best rooting medium is 1/2WPM,the rooting index is 284.81.In this study,we determined the best method of sterilization of Salix,selected the best lateral bud induction medium and the best rooting medium of eight species of Salix,and established the asparagus aseptic system.It is helpful for the establishment of Salix micropropagation system and study on virus-free seedlings.And it is conducive to the realization of Salix factory nursery. |
