Anaplasmosis of bovine is a kind of blood-borne disease caused by the spread of arthropod,such as ticks,which causes fever,emaciation and jaundice.The disease is a worldwide distribution,and is listed as one of the major diseases of animal quarantine by the world organization for animal health(OIE).Especially in the areas of severe epidemic of ticks,which causes the high incidence of livestock and a significant economic loss.There are many studies on the prevalence and the genetic characteristics of the disease at home and abroad,but there are few reports about the prevalence of the disease and the type of pathogens in the upper reaches of the Tarim river basin.The aim of this study is to know the infections with anaplasmosis,pathogen species and analysis of its molecular genetic characteristics in order to provide some basic data and supports of diagnosis,treatment and prevention on anaplasmosis of bovine in the industry.In this study,the method of PCR(conventional PCR and nested PCR)was used to detect the non-pulp type of dairy cows infected in the upper reaches of Tarim River,and 17(city,county)Region 488 samples of dairy cow blood samples were examined.The results showed that the total infection rate of non-pulp was3.5%(17/488),Anaplasma bovis,A.phagocytophilum,A.marginale infection rate 2.6%(13/488),2.9%(14/488)and 0.4%(2/488),respectively.There were 12 samples of bovine no pulp and phagocytic cells without pulp mixed infection,the infection rate was 2.5%(12/488).In the upper reaches of Tarim River,the infection rate of cows anaplasma is low.In order to study the molecular genetic characteristics about Cow derived Anaplasma in the upper reaches of Tarim River,PCR test results for the positive samples were sent to sequencing,Based on the A.bovies r RNA gene loci,13 samples were sequenced successfully,Based on the A.phagocytophilum16 S r RNA gene locus,13 samples were successfully sequenced,Sequencing 2 samples Sequence Based on MSP4 Gene of A.marginale sequencing were unsuccessful.Results show,In the A.bovis 16 S r RNA gene locus,only 1 of the 13 sequences were A.bovis,sharing 100% homology to cattle-derived A.bovies isolates from Henan Province in China(Gen Bank accession no.KX450273),The other 12 are A.platys,sharing100% homology to sika deer-derived A.blatys isolates from China(Gen Bank accession no.KJ659044).16 S r RNA gene locus based on A.phagocytophilum,sharing 100% homology to sheep-derived A.phagocytophilum isolates from Xinjiang in China(KJ782381),sharing 100% homology to cattle-derived A.phagocytophilum isolates from Xinjiang in China(KJ782390).haring 99.9% homology to species indeterminate Bactrian camels-derived isolates from Gansu Province in China(KU554739),only one base difference.The results show that,Distribution characteristics of genetic diversity of Cow derived Anaplasma in upper reaches of Tarim River Basin,A.phagocytophilum and A.platys are zoonotic species,Risk of zoonotic disease.In this study,PCR method were used to detect the blood samples of cow in the upper reaches of Tarim River.The results showed that the dairy cows had no pulp infection rate,but there were genetic diversity distribution.And flat no pulp for the dominant species,there is a zoonotic risk.This study provides a reference for the propagation,diagnosis and prevention of no pulp disease,which lays the foundation for further research on the monitoring,immunology and public health of no pulp disease.
