Anaplasmosis is an infectious disease by arthropod with Anaplasma to spread.Anaplasma belongs to Rickettsiales,Anaplasmataceae,Anaplasma.The main study of this type was Anaplasma ovis,Anaplasma phagocytophilum,Anaplasma bovis.After sheep infection that it has weight loss,anemia,jaundice and other clinical symptoms that leading to a decrease in weight gain and a decrease in forage utilization,which can lead to serious death and serious economic losses.The disease mainly through ticks and other media transmission,because of a wide range of ticks and a wide range of activities in the pastoral areas,the disease need to conduct long-term monitoring and prevention and control.In order to understand the infection of Anaplasma disease in sheep of Kashi area in southern Xinjiang,a total of 278 sheep blood samples were examined in 12 areas of Kashi by Giemsa staining and microscopic observation.The results showed that Anaplasma positive samples were 126 and the total infection rate of Anaplasma was 45.3%.Anaplasma of sheep of the infection rate was significantly different(χ2=35.7,p<0.01)by statistics in different sampling locations.Infection rate of Anaplasma(46.6%)in scattered sheep was higher than Infection rate of Anaplasma(40.7%)in captive sheep,but it is not significant difference about infection rates of Anaplasma(χ2=0.7,p>0.05)in sheep in different farming patterns.The survey results show that the Kashi region of sheep Anaplasma infection is serious that should strengthen the monitoring and prevention of the disease.For a more accurate understanding of South Xinjiang in Anaplasmosis of sheep of popular situation,a total of 637 sheep blood samples from 2,523 sheep whole blood were collected from 21 regions of Xinjiang South Xinjiang.Nested PCR was used to detect Anaplasma ovis in sheep blood DNA by the MSP4 gene.Nested PCR was used to detect Anaplasma phagocytophilum and Anaplasma bovis in sheep blood by 16 S r RNA gene.527 samples were infected by Anaplasma,and the total infection rate of Anaplasma was 82.7%.There are Anaplasma ovis,Anaplasma phagocytophilum,Anaplasma bovis three kinds of Anaplasma infected sheep blood,the infection rates were 72.2%,20.3% and 52.9%,the results show that the dominant species of Anaplasma in Xinjiang is Anaplasma ovis.Mixed infection rate of Anaplasma is common and the mixed infection rate was 50.5%.Infection rate of Anaplasma(83.7%)in scattered sheep was higher than Infection rate of Anaplasma(78.4%)in captive sheep.Anaplasma of sheep of the infection rate was significantly different(χ2=122.5,p<0.01)by statistics in different sampling locations.It is not significant difference about infection rates of Anaplasma(χ2=2.3,p>0.05)in sheep in different farming patterns.The results showed that the infection situation of Anaplasma in southern Xinjiang was generally popular,and we should strengthen integrated control to Anaplasmosis.This survey provides the basis for the prevention and control of Anaplasma disease in the southern Xinjiang region.Sequencing of partial results was taken from the samples that were positive for PCR,analyze the sequencing results and establish a phylogenetic tree.The results showed that the partial sequence had a base difference from the reference sequence,and the rest of the sequence was identical to the consensus sequence.Through the establishment of phylogenetic tree,we can see that Anaplasma ovis,Anaplasma bovis,Anaplasma phagocytophilum have no geographical isolation and differentiation,and Anaplasma phagocytophilum has host specificity.Although there are many studies of Anaplasma,there are few reports on the epidemic and pathogenic species of Anaplasmosis in the southern Xinjiang region.Through this study,we can see the epidemiological situation of Anaplasma in Xinjiang and the species of Anaplasma in sheep,and analyze its genetic characteristics to judge the genetic diversity and host adaptability of Anaplasma.So as to lay the foundation for the prevention and treatment of sheep Anaplasmosis,which greatly reduces the harm caused by the disease to animal husbandry in southern Xinjiang.
