Objective With the indexs of polygala fingerprint chromatographic peak area and the peak area ratio of tenuifolia, polygalaxanthone III and acyl sucrose content of 3, 6’-disinapoylsucrose, biological effect of nerves, biological effect of intelligence, based on the fuzzy mathematics and AHP the comprehensive evaluation model on the quality of hemp is constructed.Methods(1) Using HPLC-UV method to analyze the 40 different medicinal materials of Polygalae Radix,establish a Polygalae Radix total extract fingerprint, measure the acyl sourose content of tenuifolin, Polygalaxanthone III and3, 6’-disinapoylsucrose. According to the similarity analysis and the principal component analysis, the evaluation of Polygalae Radix can be concluded.(2)Regard the average sleep time by pentobarbital sodium in mice sleep experiment and the number of errors during the mice jumping test as standard, to establish the detection methods of Polygalae Radix biological effect of nerves and biological effect of intelligence.(3)Set up an evaluation factor set which including the evaluation factors of biological effect of nerves,biological effect of intelligence, index component content and fingerprint chromatographic peak area and the peak area ratio, take the Polygalae Radix from different areas as the object, establish a single factor evaluation and quality matrix of Polygala tenuifolia and make a comprehensive evaluation. Using the AHP to determine the weight of evaluation factors, build a model of Polygalae Radix comprehensive evaluation on quality, and calculate the values of quality characteristics of Polygalae Radix from different areas.ResultsThe content detection shows that the average content of tenuifolin is 3.568%,of Polygalaxanthone III is 0.225%,and 0.598%of 3, 6’-Disinapoylsucrose. The analysis of fingerprint common peak shows that peak 6 3, 6’-Disinapoylsucrose has the largest average peak area, which is 12029.15. The second one which has the largest common peak area is peak 10 with the area of 5428.96, the peak 6 3, 6’ account for the 40.78% of all the Polygala tenuifolia common peaks of 40 samples. The similarity results show that among 40 different medicinal materials of Polygala tenuifolia, the similarity stays between 0.636~0.951, which means that different medical materials from different region has certain differences and different medical materials from the same region also have some kind of differences. These may be related to different germplasm,cultivation and different recovery time.This thesis establishes a new method to detect biological effect of nerves and biological effect of intelligence which adopt the reaction of parallel lines(2, 2) method. According to the analysis of the biological effect of nerves and biological effect of intelligence of different medical materials from different regions show that the biological affect of nerves achieves the maximum 181.45U/g in Xia County, Shan Xi Province and the minimum 64.89U/g in Neiqiu, Hebei Province. The average is 112.80 U/g. The biological effect of intelligence achieves the maximum 180.31U/g in Suide, Shanxi Province and the lowest is 63.30 U/g in Nianzhang, Shanxi Province, the average is 119.0 9U/g.The preliminary establishment of a comprehensive evaluation of Polygalae Radix quality model is achieved, and calculates the quality characteristics of 40 different values of Polygalae Radix(TQI), as a result, Shanxi Xinjiang Yangwang TQI to a maximum value of 0.6982, followed by Shanxi, Yicheng 0.6573, the Hebei Neiqiu is the lowest 0.1350.There are 9 regions’ the TQI values reach above 0.5; 26 samples’ TQI value in most regions between are 0.3-0.5, the other 5 regions the TQI value are below 0.3.ConclusionThe thesis combine the sedative effect of Polygalae Radix, intelligence effect, fingerprint and the content of index components together, using fuzzy mathematical principle of analytic hierarchy process(AHP), the initial establishment of comprehensive evaluation model for characterization of Polygalae Radix quality is built, solve the problem of the single index evaluation of Polygalae Radix quality, which provides some ideas for the study of comprehensive evaluation of Chinese traditional medicine genuine quality.
