The seeds,rootstocks,scions and different grafting combinations of different Chang-lin cultivars of Camellia oleifera were chosen as experimental materials.The difference and variation of morphological index of seeds and rootstocks were studied and the physiological and biochemical indexes of different grafting combinations were compared and analyzed to provide the basis for breeding good varieties of rootstocks,optimizing the grafting combinations and improving the survival rate of nurse seed graft.The results were shown as follows:1.the morphological characteristics of seeds and stocks of ten Camellia oleifera cultivars(Chang-lin3#,4#,18#,23#,27#,40#,53#and166#)were studied.Results showed that extremely significant differences were existed in morphological characteristics of the seeds and the rootstocks.The maximum totality variation of morphological characters of seeds was single quality with the value of 0.12~4.19 g(CV=38.18%),then the following order were hilum length with the value of 0.84~10.56 mm(CV=28.25%),hilum width 0.61~7.50 mm(CV=23.17%),7.81~78.3 mm(CV=22.09%),seed thickness 1.36~20.24 mm(CV=21.47%),seed length-width ratio 1.00~6.11(CV=21.03%)and seed width 6.04~23.47 mm(CV=17.48%).The maximum totality variation of morphological characters of rootstocks was epicotyl length 0~19.00 cm(CV= 35.49%),the following order were epicotyl diameter 0~17.30 mm(CV= 34.45%),hypocotyl diameter 0~7.60 mm(CV= 25.26%)and hypocotyl length 0~19.50 cm(CV= 19.87%).Highly significant correlations were observed in seed weight,seed lengh,seed thickness,epicotyl length,hypocotyl length,epicotyl diameter and hypocotyl diameter(except there was no significant correlations between epicotyl length and seed lengh,seed thickness.There was significant correlations in epicotyl diameter and hypocotyl diameter.)2.Ten seeds of different Chang-lin cultivars were divided into three types according to seed weight(Chang-lin3#,4#,18#,21#,23#,27#,40#,53#,55#and166#).The weight,epicotyls length,hypocotyls length,epicotyls diameter,hypocotyls diameter of seed of different Camellia oleifera cultivars were studied.Results showed that Chang-Lin18# performed stronger than the other nine cultivars at the indexes level.On the contrary,chang-lin23# performed the weakest.3.The interaction between scions of Chang-lin3#,4#,18#,23#,27#,40#,53# and166# and rootstocks of different grafting combinations of Camellia oleifera were studied.The results showed that the grafted survival rate of Chang-lin 18# was lower than that of the other scions.A strong correlation existed in the different scions combinations with the same stock,which indicated that the genetic characteristics of the scions affected the growth of the rootstock.4.The root system of the grafted seedling with the scions of Chang-lin 3#,4#,18#,23#,27#,40#,53#and166# and the stocks of Camellia oleifera were determined.Results showed that the scion of different cultivars have an significant influence on the root system.The results of the experiment on grafted seedings of 8 C.oleifera cultivars showed that significant differences were existed in root volume,root surface,root length,root biomass and other indicators after two years' growth.The maximum total length of roots was 3 times longer than the minimum with the value of 1718.04 cm.The largest root surface area was 3.1 times larger than the smallest with the value of 498.27 cm2.The maximum root volume was 3.2 times larger than the minimum with the value of 12.16 cm3.In addition to a few exceptions,there was significant correlation between each index.Compared with different root levels,the root length of 0.0 < d < = 0.5 mm was the longest,but the root surface area and root volume was minimal.The root length of d > 2.0 mm was the shortest,root surface area and root volume was the biggest.5.Taking Chang-lin 18# for rootstock and Chang-lin 4#,18# for scion,using nurse seed graft method,the grafting affinity for physiology through the mortality in the process of grafted combination development and its related physical and chemical indexes were determined and analyzed.Results showed that the conclusions of 0h-24 h and 1d-55 d were the same,and the change trend of physiological indexes of two graft combinations were basically the same except soluble sugar content.The SOD activity and MDA content in the combination with a good affinity were lower than that in the combination with a poor affinity.The POD activity and soluble protein in the combination with a good affinity was higher than that in the combination with a poor affinity;the peak of SOD activity,POD activity and MDA content appeared earlier in the combination with a better affinity than that in the combination with a poor affinity.The affinity of two combinations had on certain relations with the CAT activity and the relationship between the affinity and the soluble sugar content of two combinations need another further study.The IAA and ABA content of 18+4 was lower than that in 18+18 in the early graft healing.But the IAA and ABA content of 18+4 was higher than that in 18+18 in the late graft healing;the ZR and GA content of 18+4 was lower than that in 18+18 in the whole stage.The ABA content and ABA/IAA ratio had an important impact on the process of graft healing.
