In this paper,the oil extraction technology of camellia seeds at low temperature was studied,and optimized by using quadratic orthogonal rotating design method.Extract protein from low temperature extracting camellia seeds-cake,and use enzyme to hydrolyze it.Influence factors of peptides molecular weight range and yield in proteinic penzymic hydrolysates were studied.Small molecular weights of peptides were produced by microwave heating and compound enzyme method,and the functional properties of small molecular weight peptides were studied.The main conclusions are:1 The extraction of camellia seed oil by low temperature methodThrough the quadratic regression orthogonal rotating combination experimental design,the camellia seed oil extraction technology by low temperature method was studied.Relevant regression models were set up:The dynamic equation of yield and extraction parameter is:Y=86.13+8.69*X1+4.90*X2+6.95*X3-9.17*X12-5.31*X22-3.17*X32-1.40*X,X2-2.91*X1X3-3.28*X2X3.The best extraction condition is:at the temperature of-10 ?,adding 120 mL anhydrous alcohol,and extracting 75 min.The actual extraction rate is 90.01%.The dynamic equation of squalene content and extraction parameter is:Y= 11.98+1.72*X1+2.10*X2-2.13*X3.The verification test and the response surface analysis shows that:besides the amount of entrainment agents,extraction temperature and extraction time,there are also some other factors.So the equation cannot simulate the process of squalene extraction very well.The dynamic equation of Vitamin E content and extraction parameter is:Y=429.40+43.54*X1+25.08*X2+36.17*X3-44.78*X12-26.17*X22-14.64*X32-6.97*X1X2-13.12*X1X3-14.14*X2X3.The best extraction condition is:at the temperature of-10 ?,adding 120 mL anhydrous alcohol,and extracting 75 min.The actual content of vitamin E is 450.32 ?g·g-1.2 The preparation of peptides from camellia seeds-cakeBy investigating the effect of hydrolysis time,hydrolysis temperature,pH,enzyme concentration,heating mode and the type of enzyme on hydrolysis degree,peptide concentration and peptide molecular weight range,the results showed that hydrolysis time,hydrolysis temperature,pH,enzyme concentration and heating mode only affect the hydrolysis degree and peptides concentration,but hardly affect the enzyme action site in the protein chain,so the enzyme molecular weight ranges are approximately the same in the enzymatic hydrolyzates Different kinds of enzymes have different optimum hydrolysis conditions,and different substrate action sites,so the degree,efficiency and product of hydrolysis are different,also the hydrolysis degree,peptides concentration and peptide molecular weight ranges are different.In those examined factors,only the kinds of enzyme can control peptide molecular weight range.3 Study on preparation of camellia seed peptide by microwave and compound enzyme method and the functional properties of peptide.The optimal technology conditions of preparation of camellia seed peptide by microwave and compound enzyme are as follows:substrate concentration 5%,first adjust pH to 10.0,then add 6000 u-g-1 alkali protease,under the 45 ? microwave conditions decompose it 12 minutes,and then adjust pH to 7.5,add 5000 u·g-1 neutral protease,under the 50 ? microwave conditions decompose it 12 minutes.until the hydrolysis degree of solution liquid reaches 23.18%,The peptides obtained belong to small molecular weight ones,and the their molecular weights mainly concentrate around 4.5 kD.Compared with the crude protein from camilla seed,the peptides produced under the optimum conditions have better water holding capacity,water solubility,emulsibility and emulsion stability and less influence from pH and electrolytes.Meanwhile,they have stronger DPPH free radical scavenging capacity than Vc and rutin,so they can be used in the food industry,medical and health care products.
