| The present study was to evaluate the effect of storage time,tenderness,fermentation on the characterization of Qingzhuan dark tea.Residual rate,intestinal propulsive rate,and the variation of intestinal microflora were determined to evaluated the influence of Qingzhuan dark tea on gastrointestinal function in mice.1.Characterization of Qingzhuan dark teaQingzhuan dark tea produced in 2010 had the highest content of free amino acids,water extract,tea polyphenols,caffeine,tea pigment,and organic acid,which may be related to the difference of raw material and process related pile.Qingzhuan dark tea produced by simple bud dark brick tea had higher content of free amino acids,tea polyphenols,caffeine,theabrownine,and organic acids,while the content of water extract,soluble sugar,theaflavins,thearubigins,and catechin was significant higher produced by two or three leaves of a bud.Qingzhuan dark tea produced in different fermentation process also varied in composition.Generally,the content of free amino acid,tea polyphenol,water extract,soluble sugar,catechin components,and organic acids were significantly decreased with increasing the degree of fermentation,while the content of caffeine and theabrownin were significantly increased.2.Influence of dose on gastric emptying,intestinal propulsion,and intestinal microflora of Qingzhuan dark tea in miceExtracts of Qingzhuan dark tea at 8.34,16.67,and 33.34 mg/10 g body weight were administered intragastrically to mice for 15 days.Results showed that Qingzhuan dark tea extracts had positive effect on gastrointestinal function in mice.Qingzhuan dark tea extracts treatment at middle dose(16.67mg/10 g.bw)exhibited the maximum effect on gastric emptying that means the dose of Qingzhuan dark tea extracts was not the higher the better on gastrointestinal function.3.Influence of storage time on gastric emptying,intestinal propulsion,and intestinal microflora of Qingzhuan dark tea in mice.Qingzhuan dark tea produced in 2016,2010,and 2006 were extracted and administered intragastrically at middle dose to mice for 15 days.Results showed that Qingzhuan dark tea extracts could reduce the gastric residual rate,promote gastric emptying and intestinal peristalsis,and increase intestinal propulsion.The results of intestinal flora showed that Qingzhuan dark tea extracts also improved the microecological environment in the gastrointestinal tract with the number of bifidobacteria and lactobacillus significantly increased and the number of enterobacteriaceae and enterococci significantly decreased.The positive effect on gastrointestinal tract of Qingzhuan dark tea showed a positive correlation with the storage time.The improvement of the gastrointestinal tract in mice of Qingzhuan dark tea was more effective with prolonged storage time and Qingzhuan dark tea produced in 2006 showed the best effect in our study.4.Influence of tenderness on gastric emptying,intestinal propulsion,and intestinal microflora of Qingzhuan dark tea in miceQingzhuan dark tea produced by different tenderness of fresh leaves were extracted and administered intragastrically at middle dose to mice for 15 days.Results showed that Qingzhuan dark tea could promote the gastric emptying and small intestinal peristalsis in mice and improve the microecological environment in the gastrointestinal tract with increasing the number of bifidobacteria and lactobacillus significantly.The effect of Qingzhuan dark tea on gastric emptying,small intestinal propulsion,and intestinal microflora are consistently with the same order: single bud,a bud and two leaves,and a bud and four leaves.5.Influence of fermentation on gastric emptying,intestinal propulsion,and intestinal microflora of Qingzhuan dark tea in miceQingzhuan dark tea produced by different fermentation process were extracted and administered intragastrically at middle dose to mice for 15 days.Results showed that mild and moderate fermented Qingzhuan dark tea could promote gastric emptying and intestinal peristalsis in mice,and non-fermented Qingzhuan dark tea did not show that effects.The results of intestinal flora showed that mild and moderate fermented Qingzhuan dark tea extracts could increase the number of bifidobacteria and lactobacillus significantly and decrease the number of enterobacteriaceae and enterococci significantly compared with non-fermented Qingzhuan dark tea.The influence of fermentation on gastric emptying and small intestinal propulsion of Qingzhuan dark tea were basically consistently with the effect on intestinal flora. |
