| With social and economic growth,people’s living level have increased.Health problems have become increased such as obesity and diabetes caused by high-energy diets.Obesity and type II diabetes(T2D)can lead to the endotoxemia due to the destruction of intestinal barrier.and it can initiate intestinal inflammation.Energy-restricting including intermittent fasting and calorie restriction is a new dietary model for managing weight.Recently Studies have shown that can reduce T2 D.However,the protective mechanism of intestinal barrier function is rarely reported.Therefore,this experiment was designed to research that intermittent fasting improve the intestinal barrier function and regulating intestinal microorganism in db/db mice.The main research content is as follows:(1)The db/db mice were divided equally,and db-m were used as control.28-day short-term intermittent fasting were used in db/db-IF groups.The study found that intermittent fasting can inhibit body weight gain in db/db mice,increase serum HDL-C,decrease serum LDL-C,down-regulate serum LPS levels,increase insulin sensitivity,and down-regulate m RNA expression of intestinal inflammatory factors.At the same time,the expression of anti-oxidation protein m RNA was up-regulated.(2)To explore the effect of intermittent fasting on the intestinal barrier function in db/db mice.H&E staining showed that intermittent fasting could effectively improve the intestinal villi damage;ussing chamber showed that intestinal permeability was decreased by intermittent fasting;Immunofluorescence assay and RT-PCR show that intermittent fasting can upregulate the expression of intestinal tight junction protein in db/db mice.(3)To explore the relationship between intermittent fasting and gut microbiota in db/db mice.It was found that intermittent fasting could improve the diversity and richness of intestinal microflora in mice.Intermittent fasting could change the composition of OTU and reduce the F/B ratio in db/db mice.It could promote relative abundance of probiotics Lactobacillus;inhibite the abundance of harmful microbiota.At the same time,the content of short chain fatty acids which were tested in feces was significantly reduced in db/db group,and intermittent fasting could increase them and improve the normal physiological function.This experiment was revealed the mechanism of intermittent fasting on the intestinal barrier function and intestinal microbiota changes and the effect of intermittent fasting on the improvement of intestinal inflammation which caused by T2 D.It can provide a theoretical basis for the application of new dietary patterns.. |
