Bottom sediments from bodies of water carried valuable information on biogeochemical processes and climatic changes.A number of chemical elements and compounds, among which sulfur occupies an important place, are used for characterising the state of bottom sediments, the mechanisms and conditions of their accumulation. There are miscellaneous test methods of sulfur in the atmosphere and water body but there are still more uncertain factors in sulfur conversion ofanaerobic environment, such as sulfide and thiosulfate, sulfite, sulfate and elemental sulfur).Due to uncertainty in the form of sulfur transformation, many limitation and restriction are found in the accuracy, reliability of surfur measurement. In this thesis, around all kinds of sulfur components?including sulfide, thiosulfate, sulfite, sulfate and sulfur etc.?are applied in the speciation analysis and quantitative tests. We use their detection methods at the same time, the independent design of a variety of detection methods(S0,SO32-and S2O32-),which summed up the extraction of various different forms of sulfur and test methods using the method of extraction of elemental sulfur,to detect dissolved sulfur by HPLC.DTNB are used as derivative agent in thiosulfate and sulfite analysis of sediments.from sulfite reducing bacteria?SRB?in acid salt anaerobic reactor.Combined with other sulfur components, the concentration changes of sulfate products were analyzed. The previous test methods in sulfate reducing product are of great error and boresome work as well. From the previous methods(detection of SO42-,SO32-and S2O32-and S0 and S2-),some novel methods(S0 and SO32-and S2O32-)are well designed by us.Sulfate is the general international method, there are some errors, but their errors are within the acceptable range,of the data is reliable. In the API standard medium,we use control variable method to cultivate the three sets of samples, one of the most representative samples is chosen to test and carry out analysis on the content of sulphate reduction product. The following results are obtained:?1?In the comparative experiment, based on the selection of extraction agent of efficient sulfur extraction, acetone is thought as the optimum one after a 10-minute ultrasonic extraction.?2?A detection limit on elemental sulfur of 1.25 ×10-3 mg/L(2.5 ×10-3 mg/kg)and detection limit of 4.18 ×10-3 mg/L(8.36 ×10-3 mg/kg)are obtained when elemental sulfur is determinated by high performance liquid chromatography?HPLC?method, meanwhile the standard addition recovery rate ranges between 100% and 120%.The result is satisfactory, since the sulfide concentration in anaerobic fermentation product are included in high performance liquid chromatography detection range,so this method is feasible to characterize the elemental sulfur of fermented product.?3?In thiosulfate and sulfite determination method,DTNP is chosen as the derivatization reagent by using multi-step gradient elution mode, which can get better test results. The detection limits of is thiosulfate and sulfite are both 1.25×10-3mg/L.The recovery rate of the thiosulfate and sulfite determinated by high performance liquid chromatography is around 100%.?4?In SRB degradation reactor, after test,it was found that the relative concentration of sulfate reduction, such as thiosulfate and sulfate is only an average of0.16 mg/L and almost negligible. All sulfur contents can be arranged in the order of S2->S0 >H2S >SO32-/(S2O32-).Among them, about 70% of the product is in sulfide form,10% is the existence of sulfur form, the rest of the sulfur is SO32-,S2O32-,S2O52-,S2O42-and other forms. In the reactor addition with goethite,a 17.2% sulfur loss was found, mainly in the form of FeS,FeS2 and organic sulfur loss.
