In recent years, Food safety has been the concern problem of the people, With the improvement of living standard and life rhythm of speeding, People pay more attention to the quality of life, more and more health food have appeared peoples vision,Phosphodiesterase type 5 inhibitors exist in anti-fatigue and aphrodisiac functional health food, if we eat the medicine that adding phosphodiesterase type 5 inhibitors all the time,there will be a lot of side effects, such as headache, dizziness, indigestion, rashes. Therefore,it is important to establish a method of high sensitivity detecting of phosphodiesterase type 5 inhibitors. Carbon nanotubes with large specific surface area, ? electrons from, van der Waals force, this article chooses carboxyl carbon nanotubes as solid phase extraction of adsorption material. Establishing a method detecting phosphodiesterase type 5 inhibitors in health food that use solid phase extraction and high performance liquid chromatography(HPLC).Firstly, this experiment optimized the chromatographic separation conditions. The best condition of high performance liquid chromatography (HPLC), ODS C18 chromatographic column (250 mm×4.6 mm,5 ?m), Working temperature is 30?, injection volume is 20 ?L.Ammonium acetate solution of 50 mM as a mobile phase, Acetonitrile and acetic acid ratio of 1:1 as B mobile phase, In this conditions, six targets the peak type sharp symmetry, good separating degree.Secondly, through single factor variable test, the parameters affect extraction efficiency were optimize, confirm the best solid phase extraction contitions, the samble volume was 25 mL, the sample flow rate: 0.8mLmin-1, eluent solutions acetonitrile/acetic/ hydrochloric acid, (9:7:0.01v/v/v). eluent volume: 6.0mL. Under the optimum conditions, the limits of detection (S/N=3) of the method ranged from0.3-0.6 ?g L-1, Six kinds of phosphodiesterase inhibitor have good linearity (R2 0.99-0.9969) within 1-50?g L-1.The mixed standard solution concentration of 10 ?g L-1 was determinate 9 times repeatly.the relative standard deviation for nine paralleled determination were below 6.91%.The contents of phosphodiesterase type 5 inhibitors in samples were measured by establishing method.The result showed that there were different kinds of phosphodiesterase inhibitors in three kinds of actual samples,Recoveries of the spiked sample (0.025?0.125?0.625 mg kg-1) were achieved ranging from 73.34-115.76%, the relative standard deviation were below 9.97 %. The method can be used in the detection of complex actual samples.
