In recent years,application of plant tissue culture technology is widely used in scientificresearch and production fields.But because of plant tissue culture technology for strict aseptic environment,resulting in higher cost,limit the development and the promotion and popularization of plant tissue culture technology bottlenecks,there is an urgent need for research and promotion.Broke the open tissue cultureof the traditional concept of aseptic culture,give high pressure sterilization and clean benches,operating out of the sterile environment.This test with red bulbs formaterials,by filtering the bacteri ostatic agent,get the right type and concentrationof antimicrobial agent,preliminary study on tissue culture of Szentendre open,optimize the rooting medium for proliferation,proliferation of best rooting concentration of antimicrobial agent,and detect open proliferat ion seedling physiological and biochemical index of red,with a view to improving the Szentendre open tissueculture system,reducing cost of tissue culture.The results were as follows:1.Through natural fell bacteria and artificial received bacteria test determine different inhibition bacteria agent of inhibition bacteria effect,results showed that:Daisen manganese zinc,and hypochlorite sodium and H2O2 has obviously of inhi bition bacteria effect,determine inhibition bacteria effective concentration hypochlo rite sodium for 0.01%-0.05% Zhijian,Daisen manganese zinc and H2O2 effective inhibition bacteria concentration for 0.05%-0.1% Zhijian,but H2O2 nature not stable,dang concentration is greater than 0.1% Shi medium within appeared large bubble,led to medium not solidification.2.Adoption of different concentrations of sodium hypochlorite and mancozeb onvac cination and disinfection,sterilization of inoculation with the traditional burningof apparatus for contrast.Results showed that higher pollution concentration of sodium hypochlorite is smaller,and the concentration of sodium hypochlorite at0.8% with traditional disinfection rate were not significant,pollution rate is only 3.78%;bycontrast mancozeb less effective,pollution rate in 1% concentrations of mancozeb were still 9.75%.Therefore,the concentration of selected 0.8% Szentendretissue culture inoculation apparatus of the so dium hypochlorite disinfection.3.The use of sodium hypochlorite and mancozeb disinfection of explants of red,results show that treatment can reduce the rate of Browning using low concentration,high concentration of disinfection can reduce the rate of pollution,considering thepollution rate,Browning rate and survival rate,select Add 0.01% medium and explant with 1% sodium hypochlorite disinfection 20 min.4.Through on red proliferation and health root medium best hypochlorite sodium added concentration for test,results showed that: hypochlorite sodium concent ration for 0.013% Shi,value-added coefficient highest,low hypochlorite sodium concentration Xia proliferation pollution rate significantly above high concentration hypochlorite sodium;Dang hypochlorite sodium concentration for 0.016% Shi,health root rate highest,hypochlorite sodium concentration high adverse yushandandan indefinite root of growth,and on health root number and root long are has effect.Therefore,considering the training targets,select 0.014% the concentration of sodium hypochlorite as red on proliferation and rooting medium for antimicrobial agents,the most appropriate.5.To save the contaminated tissue culture seedlings of traditional sterilization,results show that longer soaking time,lower the pollution rate,but a high concentration of sodium hypochlorite on explant toxicity is serious,leading to low survival rate.Low concent ration,long soak can be effective in reducing pollution,increasing pollution plantlet survival rate,soak 5-10 min inoculation,to add 0.4% sodium hypochlorite on the medium can save nearly 60% of the contaminated tissue culture seedlings,losses caused by pollution.6.Through red traditional and open proliferation training group culture seedlings for material,compared two species group culture way on red physiological biochemical index of effect,results showed that:(1)hypochlorite sodium of joined on plant produced stress role,to adapted adversity,soluble sugar content increased and regulation penetration pressure,adapted outside stress,indefinite bud into medium early,not adapted new environment,consumption itself can dissolved sugar,for growth differentiation,grew recovery Hou began
