The cells of heart tissues derivedfrom Chinemys reevsii Gray were cultured in vitro with the medium 199 supplemented with 20%calf serum.The results of preliminary culture showed there are three types cells migrated out from the heart tissues.They are respectively suspended blood cells,attached epithelioid cells and attached fibroblast-like cells.At post-inoculation,the blood cells first migrated out from the tissues.At 3 d post-inoculation,epithelioid and fibroblast-like cells began to migrate out from the tissues.Being cultured for 14 days,it was formed some great piece cell cluster compositive with epithelioid and fibroblast-like cells,respectively. The present study indicated that the cells could change shape in the cultured process, the same tissue could form different morphous cells and the same cell population could as well as form different morphous cells.A great deal cells have proliferated and some cells were bankrupt in contact inhibition in the late phase of preliminary culture.The study showed that the heart cells of preliminary culture have step by step achieved transformation and formed the cell population possessed of proliferation ability.The cells of foot and mantle tissues derived from Ampullaria gigas Spix were cultured in vitro with the modified medium 199.At 6h post-inoculation,cells began to migrate out from the tissues.At 3 d post-inoculation,the outgrowth was formed around the tissues.Being cultured for 21-28 days,most of the bottom of the culture flask was covered by the cells.The cells of foot and mantle tissues have been subcultured to the eighth and ninth generation,respectively.There are two types of cells in the preliminary cell culture of the foot and mantle tissues.One was small epithelioid cells,which were rounded,elliptic or polygonal with diameter of 7-15μm; the other was middle epithelioid size and large cells,which is elliptic and polygonal with diameter 15-30μm.There were plenty of the small epithelioid cells,which proliferated faster and could form cell monolayer in the preliminary culture and subculture.The results of preliminary culture and subculture showed that the growth ability and adherence of mantle tissue cells was better than that of foot tissue ones. The result of Hoechst fluorescein stain revealed that the apoptosis index of mantle tissue cells was apparently less than that of foot tissue cells.The present study implied that mantle tissue could have potential to be established as a continuous cell line.
