Tissue Culture Of Six Bryophytes

In this study,the sphagnum,broad leaf sphagnum peat moss,egg moss,fine rib thin beak moss,moss blond and brown Folioceros fuciformis were used as reaserch materials.Here,we provided the basic data and strategies for cultivation of bryophyte tissues,by study the process of disinfection and optimization of culture mediums,respectively.The main results of this study are as follows:(1)Disinfection were carried by 75% alcohol and sodium hypochlorite,the four kinds of media were the basic medium of Knop,Beneck,BG-11 and MS.Screening in the different sucrose concentration: The condition of disinfected the three variety of multi-pattern peat moss from Sphagnum Ehrh,egg Leaves of peat moss and mossy peat moss were used 200mg/ml penicillin in 2 to 2.5 h,75% alcohol in 60 s and 0.2%-0.3% sodium hypochlorite in 60 s.The optimum culture medium for the highest growth rate of buds was Beneck medium with 2.5% sucrose concentration.The best concentration of sucrose in the Knop culture was 2% for peat moss cultivation.(2)The disinfectant methods of polytrichum commune were immersed in 50 mg/L penicillin for 3 h,75% alcohol for 60 s,and 0.4% sodium hypochlorite for 90 s.After 5 min of ultraviolet irradiation,the survival rate of blister meconium was 80%,and with 35% contaimination rate.The optimized concentration of sucrose was 2.5% when the hormone was not added in Beneck medium.When the NAA and 2-4 and D hormones were added,the best concentration of sucrose changed to 2%,and this was the best for the growth of the gold moss.(3)The best sterilization method of Rhynchostegiella leptoneura was immersed in 150 mg/L penicillin for 2 h,75% alcohol for 60 s,and 0.2% sodium hypochlorite for 90 s.The Ribs beak mushrooms grown better when it cultured in the basal Knop medium,however,the Ribs beak mushrooms be inhibited and easier be contaminated when added the sugar.(4)The foliage of Folioceros fuciformis plant was immersed in 150mg/mL penicillin for 2.5 h,the concentration of sodium hypochlorite was 0.15%.Under this condition,it has the lowest contamination rate and the survival rate was best.Disinfection of spore were used 100mg/mL penicillin soaked 2 h,75% alcohol soaked 30 s and 0.1% sodium hypochlorite soaked 60 s.When used UV lamp irradiation 120 s,the germination rate was best,and lower contamination rate.The best medium for spore germination and leaf growth was 1/2MS medium.Furthermore,the technique of tissue cultures also be used for the plant algae and epiphytic algae studies,and we also found that epiphytic algae was conducive for other mosses to be greener.
