Zinc and iron,calcium are essential elements for the growth and development of plants.When zinc,iron,calcium is excess or deficient,it will be detrimental to growth and development of plants.Therefore,more and more researchers focus on homeostasis mechanisms of Zn,Fe,Ca in plants.First,We obtained two loss-of-function ZNE1?Zinc Nutrition Essential1?homozygous mutants and named znel-1 and zne1-2 respectively.Under conditions of zinc excess,iron deficient,calcium deficient,the growth of the two mutants were impaired compared to Col-0?Wild type?,the leaves of mutant showed chlorisis phenotype,the root were shorter than Col-0.So zne1-1 and zne1-2 exhibited zinc excess sensitive,iron deficient sensitive and calcium deficient sensitive phenotype.We introduced functional complementation vector which contain ZNE1 to zne1-1 and zne1-2 and got two transgenic lines and found that the phenotype of transgenic plants were similar to wild type.To confirm that whether ZNE1 were involved in the absorption or accumulation of zinc,iron and calcium in plants,we measured total contents of Zn,Fe and Ca in plants by Inductively Coupled Plasma?ICP?.We found that the two mutants and Col-0 exhibited no significant different under condition of different Zn2+,Fe2+ and Ca2+concentrations.This indicated that ZNE1 gene don't participate in accumulation of Zn,Fe and Ca,but may be involved in the distribution of these elements.By Trypan Blue Staining,cell around the vascular bundle of mutants showed dark blue and were impaired.We speculate that vascular cells around the vascular accumulate more Zn2+We used GUS histochemical staining to study the tissue specificity of ZNE1 promoter.ZNE1 was expressed in roots and leaves.In this study,it was found that ZNE1 may be involved in the homeostasis mechanisms of Zn,Fe and Ca.Our research about the expression pattern and physiological function of ZNE1,and it provide the basis for further study of ZNE1 and optimizing of the contents of Zn,Fe and Ca in crops.
