Plant senescence is a series of changes in plant growth and development process triggered by avariety of factors, and it is accompanied by a series of physiological changes in the senescencing process,the most obvious of these changes is the change in chlorophyll content. With the degradation of chlorophyllin plant, chlorophyll levels is decreased, lutein and carotene content increased, thus showing the change ofleaves from green to yellow in plants, which called yellowing. This change often occurs in the early stagesof the process of senescence, this changes between chlorophyll content and plant senescence that shows anegative correlation, so some people think that changes in chlorophyll content is a sign of leaf senescence.Chlorophyll fluorescence imager can visually compare the observed changes in leaf chlorophyllcontent, according chlorophyll fluorescence parameters can be speculated that the changes in leafphotosynthesis. This experiment using chlorophyll fluorescence as filters senescence in Arabidopsismutants. With T-DNA insertion mutant seeds as screening material and chose two mutants which are namedthem f41and h1in these screened material. After identification found that they are both Both T-DNAinsertion mutants, and through genetic analysis the results shows that these two mutants are single recessivemutant gene. h1has obvious leaf yellowing phenomenon throughout the growing stage, and there aresignificant differences in chlorophyll content and soluble protein content with WT. f41chlorophyllfluorescence phenotype is significantly different from WT, after TAIL-PCR Cloning f41and use sequencecomparison, found that the mutant gene may be associated with balance and transmembrane transport ofiron, cobalt and other divalent metal ions. The analysis found that T-DNA insert it in the first frame of thefirst coding exon and the direction of T-DNA insertion and gene are in the same direction. After detection of f41-related physiological indices found that mutational gene cause changes in physiological index. Theresult of comparing with the wild type showed that the gene may be related with senescence.
