The saline-alkali soil is widely distributed in China and is an significant reserve resource,the mechanism of plant response to salt stress has been thoroughly studied in the model plan Arabidopsis thaliana.However,the effect of alkali stress on plant is less studied,mainly focus on those non-model plant and the specific mechanism of plant response to alkali stress is not clear.In this study,we selected Arabidopsis thaliana as the research plant,in order to find some genes that can regulate the alkali tolerance by forward genetic screening of mutants through the stress of Na HCO3,which is also the main component of alkali soil.During the study,we used the chemical mutagen EMS to induced about 10000 seeds and screened the M2 generation under 1/2 MS culture containing Na HCO3 stress,and chosen the sensitive phenotype plants that are with yellow leaves and curled roots as our candidate mutants.We named that as sodium bicarbonate sensitive?scs?.Then we collected samples of mutant scs46-1 to analyse its single nucleotide polymorphism through the second-generation sequencing and obtained four candidate genes.By screening the phenotypes of T-DNA insert mutants of these four candidate genes,we found that phenotype of one mutant of gene At2 g XX300 showed sensitive to Na HCO3 stress.Furthermore,overexpressed At2 g XX300 can enhance plant tolerance of Na HCO3 stress.Therefore,we preliminary determined that SCS46-1 is the gene At2 g XX300.We also collected the root samples of scs46-1 and wild-type for transcriptome sequencing in order to study the signal pathway of SCS46-1 that regulating plant Na HCO3 tolerance.Through GO and KEGG enrichment,it was found that the genes related to nitrogen metabolism,alpha-Linolenic acid metabolism,ascorbate and aldarate metabolism,arginine and proline metabolism were significantly enriched in scs46-1.Throughupon,we speculated that SCS46-1 may affect the tolerance of plant tolerance of Na HCO3 stress through the pathways above.
