The Polyphasic Taxonomy Study Of Two Novel Strains And Immobilization And Screening Of Denitrifying Microorganisms

This study mainly consists of two major parts,one is the polyphasic taxonomy study of two novel strains,and the other is immobilization and screening of denitrifying microorganisms.Two novel strains were isolated from at Mohe basin and Guizhou chishui.Through the analysises of genotic,phenotic,and chemotaxonomic to determine the taxonomic status of M0322 and GSA192.And the immobilized ball formula and the denitrifying microorganisms of efficiency were determined through embedding fixed bacterium and screening.The new species M0322 was defined as Altererythrobacter buctense,a rod-like gram-negative bacterium,no flagellar growth,no motility,facultative anaerobes.The optimum growth condition is R2A,30?,pH 7.0,0%NaCl;the main isoprenoid quinone is Q-10,the main polar lipids are phosphatidylethanolamine,phosphatidylglycerol,phosphatidylcholine,three unknown phospholipids and an unknown glycolipid,the main fatty acid type is the summed feature3(29.5%),summed feature 8(27.8%)and 11-Methyl C18:1 co7c(22%);the G+C percentage is 64.6 mol%,and the 16S rRNA gene number is KJ599648.The new species GSA192 was defined as Flavobacterium chishuiensis,a rod-like gram-negative bacterium,no flagellar growth,no motility.The optimum growth condition is R2A,28?,pH 7.0,0%NaCl;the main isoprenoid quinone is MK-6,the main polar lipids are phosphatidylethanolamine,three unknown aminolipids and an undentified lipid,the main fatty acid type is C15:0iso(12.57%),C15:0 anteiso(11.34%),summed feature 3(18.07%,C16:1?6c and/or C16 1 ?7c),C16:0(10.31%);the G+C percentage is 36.1 mol%,and the 16S rRNA gene number is KF322525.Through the study on immobilization of denitrifying microorganisms,the optimum conditons were determined,which were the 4%sodium alginate,0.5%calcium carbonate,0.5%polyvinyl alcohol,crosslinking agent for the formulation of acid +1%calcium chloride,crosslinking time 20h.The optimal amount of bacteria is 1:3,the optimum pH is 8,the optimal temperature is 30 ?.And Paracoccus versutus and Paracoccus bengalensis,two kinds of bacteria nitrification effect is better,Pscudomonas stutzeri and pscudomonas caeni denitrification effect is better.
