As China’s comprehensive national strength and the level of science and technology have been constantly improved, work related with space station construction is moving forward steadily. During the process of space station construction in the future, scientific research experiments will be gradually expanded, which included higher plants as the object of space life science experiments. Recent years the model of space life science experiments have gradually changed from early mode of detection and analysis of sample recovery to experiment in orbit, so the demand for space experiments in situ detection process has become increasingly urgent.This paper introduces the present situation of the development of multi-spectral fluorescence imaging system, and then introduces the mechanism of chlorophyll fluorescence and theoretical foundation of correlation between chlorophyll content and fluorescence. We propose a plan of dual fluorescence spectral imaging detection technology which is suitable for chlorophyll content detection during the development of higher plant, and describes the design of the light source, filter wheel and circuitry in detail. In the Part of the detector circuit system, we describe configuration register, shutter mode, read of image data, data transmission, motor drive modules and so on in detail. In the part of software design, we introduce the VHDL programming and PC software. We also measure chlorophyll fluorescence spectra at different growth stages with fiber spectrometer, and then use MATLAB to extract the spectral characteristic parameters and establish regression model. Finally, we do fluorescence imaging experiments of leaf with the system we have set up, and then use the relationship established by fitting to retrieve the profile of chlorophyll concentration in the whole leaf.
