In recent years, Surface Plasma Resonance (SPR) sensor is widely used in biological and chemical fields because of its real-time detection, fast response, high sensitivity, less samples required and without tags, etc. On the basis of that, optical fiber SPR sensor which has the advantages of high flexibility, high plasticity, low cost and easy miniaturization is of great concern. Combining with the experience of previous research, we demonstrated a high-performance fiber sensor which has a capillary optical waveguide as the sensing carrier, simulation, manufacture and biochemical detection method based on SPR principle, the specific content is as follows:First, this paper summarizes the history of SPR technology development, and elaborates the present research status and future development tendency of the SPR technology, starting from the angle of physics. On the basis, we analysis the basic principles of the optical fiber SPR sensor in detail, and develop a theoretical model of the optical fiber SPR sensory system.Secondly, we find the different factors affecting SPR phenomenon detection, according to the theory of SPR sensor and detection principle of the mathematical model. Through the Matlab software programming, by the method of controlling the variable, simulation results are calculated for the various influence factors in the theory, the ideal conditions of the influencing factors in theory are obtained, respectively.Then, on the basis of the theoretical simulation, we build the detection system of capillary optical waveguide SPR sensor, and introduce the parameters of each part of the sensory system. The paper proposes a novel carrier of sensing probe-capillary optical waveguide. We take a basic performance test of optical fiber SPR sensor system. ITO thin film is presented to improve the optical performance of sensing probe, and compares them with optical fiber SPR sensor probe. It improves the sensitivity, resolution and stability of the SPR sensor system.Finally, in order to validate the application value of the system in biochemical tests, we design two kinds of biochemical application from a practical standpoint, one is the analysis of waste water degradation process based on SPR spectrum, the other is testing the specificity of HSA, through the discussion and analysis of experimental results, we get an approving optical fiber SPR sensor system.
