Object: To investigate the expression of CD147, MMP-9, and HER-2 protein in human gastric carcinoma,and to study their relationship with the invasion, metastasis and prognosis.Methods: Immunohistochemistry method was applied to detect the expression of CD147, MMP-9 and HER-2 protein in gastric carcinoma(n=63) and the adjacent normal tissues(n=63), and the results were compared to clinical pathological factors. Univariable survival analysis and COX regression were used to predict prognosis.Results:(1)In gastric carcinoma and adjacent normal tissue the positive expression rate of CD147 protein was77.8%(49/63) and 25.4%(16/63) respectively, there was significant difference between the two groups(P<0.01). The expression of CD147 was significantly correlated with depth invasion, TNM stages, lymph node metastasis and Ki-67 expression(P <0.05), but no association was observed with the age, gender,tumor size, the degree of differentiation, pre-operation CEA and CA199 expression(P>0.05).(2)In gastric carcinoma and adjacent normal tissue the positive expression rate of MMP-9 protein was81.0%(51/63) and 23.8%(15/63) respectively, there was significant difference between the two groups(P<0.01). The expression of MMP-9 was significantly correlated with depth invasion, TNM stages, lymph node metastasis and Ki-67 expression(P <0.05), but no association was observed with the age, gender,tumor size, the degree of differentiation, pre-operation CEA and CA199 expression(P>0.05).(3)In gastric carcinoma and adjacent normal tissue the positive expression rate of HER-2 protein was34.9%(22/63) and 0%(0/63) respectively, there was significant difference between the two groups(P <0.01).The expression of HER-2 was significantly correlated with lymph node metastasis and Ki-67 expression(P<0.05), but no association was observed with the age, gender, tumor size, the degree of differentiation,depth invasion, TNM stages, pre-operation CEA and CA199 expression(P>0.05).(4)Kaplan-Meier analysis demonstrated depth invasion, TNM stages, lymph node metastasis, CD147,MMP-9 and HER-2 protein expression were significant factors influencing the survival of postoperative GC patients(P<0.05). COX regression showed that depth invasion, TNM stages, lymph node metastasis,CD147, MMP-9 and HER-2 protein expression were not independent prognostic factors for survival of postoperative GC patients(P>0.05).(5)Spearman analysis showed that CD147 protein expression was positively correlated with MMP-9protein expression(r=0.907,P=0.000). CD147 protein expression was positively correlated with HER-2protein expression(r=0.311,P=0.013). MMP-9 protein expression was positively correlated with HER-2protein expression(r=0.271,P=0.032).Conclusion:(1)CD147, MMP-9, HER-2 protein were expressed in human gastric carcinoma at different levels. CD147,MMP-9 and HER-2 may play a role in proliferation activity and invasiveness of gastric carcinoma.(2)Over-expression of CD147、MMP-9 and HER-2 were risk factors in predicting the prognosis of(3)postoperative GC patients, but they were not independent prognostic factors for survival of postoperative GC patients.(4)CD147, MMP-9 and HER-2 protein expression were closely related in gastric carcinoma, they may through certain signaling pathways synergy in the metastasis of gastric carcinoma.
