Backgrounds & ObjectivesTo explore the gastric aging has the significance in improving the life quality of the elderly and reducing the incidence rate of gastric cancer. The hunger hormone Ghrelin is an endogenous ligand of growth hormone secretagogue receptor, mainly by secreted by gastric, which has extensive biological functions. The former study has found that the expression level of Ghrelin may be related to aging, atrophic gastritis, helicobacter pylori infection and so on. Therefore, the objectives of this experiment was to study the expression of Ghrelin in normal and aging gastric mucosa, to figure out the role of Ghrelin in chronic atrophic gastritis and aging stomach.Methods50 patients with chronic atrophic gastritis and 50 young healthy controls were enrolled to the test, who all saw their doctor in GI department of Sir Run Run Shaw Hospital during the period from June 2015 to January 2016. Each of them received gastroscopy. After biopsy on gastric antrum and being pathologically confirmed by pathology department, the specimens were embedded in paraffin and sectioned. The expression of p16INK4a and Ghrelin were detected by immune-histochemistry.Results1) The expression of p16INK4a in the atrophic group is higher than in healthy control. The difference is statistically significant ((9.4± 2.7) vs (2.7± 3.3), p<0.05). In the atrophy group, In the atrophy group, the expression of p16INK4a is slightly lower in male than in female, and higher in severe atrophy than in moderate atrophy, but no statistically significant difference. The expression of p16INK4a in severe intestinal metaplasia was higher than in moderate intestinal metaplasia, and the difference was statistically significant ((8.4±2.2) vs (10.1±2.8), p<0.05).2) The expression of Ghrelin in the atrophic group is slightly higher than in healthy control but the difference is not statistically significant. In the atrophy group, the expression of Ghrelin is slightly higher in male than in female, and lower in severe atrophy than in moderate atrophy, as well as in severe intestinal metaplasia than in moderate intestinal metaplasia, but the difference is not statistically significant.Conclusions1) The expression of p16INK4a in chronic atrophic gastritis is higher than that of normal, and the expression is increased with the increase of the degree of intestinal metaplasia. Gastric mucosa of chronic atrophic gastritis can be used as a tissue specimen of aging stomach.2) The changing of Ghrelin’s expression in the aging stomach are not obvious. Ghrelin’s expression level in the aging of the stomach is still controversial, which need to be further improved.
