Objective:To evaluate the therapeutic effects of Shengwei Prescription and Hericium on chronic atrophic gastritis. To investigate the clinical effect and mechanism of Shengwei Prescription and Hericium erinaceus on chronic atrophic gastritis, to provide the oretical basis and safety index for the treatment of chronic atrophic gastritis.Methods:Clinical research:Totally 60 cases of chronic atrophic gastritis patients with spleen deficiency and blood stasis syndrome were selected according to the inclusion criteria and were randomly divided into experimental group (Shengwei Prescription+Hericium group, n=30) and control group (Shengwei Prescription, n=30). After 6 months of treatment, outcome measures, such as the clinical symptoms score, gastric mucosa atrophy, intestinal epithelial metaplasia and dysplasia were observed before and after the treatment for patients in both groups.The experimental study:90 healthy female SPD rats were selected and divided into blank group(n=15)and model group(n=75). Blank group was treated with normal diet and drinking water, while the model group with free drinking MNNG water, coordinated with hunger and satiety method to build the rat model of chronic atrophic gastritis. After modeling for 20 weeks, the successful model was cinfirmed, then the model rates were randomly divided into model group (n=15), Weifuchun group (n=15), Shengwei Prescription group (n=15), Hericium group (n=15), Shengwei Prescription group and Hericium group(n=15). The rats in blank group were continuously administrated with normal diet and drinking water;rats in model group were intragastrically administrated with equal volume of normal saline;rats in weifuchun group with equal volume of medicine weifuchun,0.39g/kg·d; rats in Hericium group with equal volume of medicine hericium granules,0.81 g/kg·d, rats in ShengweiPrescription group with equal volume of Shengwei Prescription,11.7g/kg·d, rats in Shengwei Prescription+ Hericium group with equal volume of Chinese medicine and hericium granules,12.51g/kg·d. The rates in all the groups orally administrated once a day for 9 weeks. At the end of 29th week, we killed all the rats and collected serums and stomach tissue. The gastric antrum tissue was fixed with formalin, embedded with paraffin, and submitted for morphology under light microscope. Rat pepsinogen Ⅱ、pepsinogen Ⅰ、IL-6、IL-8、 CRP、NF-κB、EGF、GAS levels were analysed by ELISA method.Results:Clinical study results show that:The effective rate of the experimental group was 100%, and the control group was 93.4%, and there was a significant difference between the two groups(P<0.05). Experimental group, the pathological effective rate was 96.7%, In the control group, the pathological efficiency was 83.3%, and there was no significant difference between the two groups(P>0.05).Significant differences were observed in total scores of clicinal symptoms between before(P<0.01). Significant difference were also observed in gastric mucosa chronic inflammation, inherent gland atrophy and intestinal metaplasia between before and after treatment for both groups, and between two groups after treatment(P<0.01 or P<0.05).Experiment results show that:(1)After modeling with MNNG freely drinking and hunger satiety dysregulation, the gastric mucosa of the rats appeared different degrees of atrophy and intestinal epithelial metaplasia. After the drug treatments, gastric mucosal atrophy and intestinal epithelium improved among rats in each group, and the effect of Shengwei Prescription +Hericium group was the most obvious. (2)ELISA results showed that:In model group, the serum levels of PGII,PGI, GAS and EGF were significantly decreased(P<0.01), but the serum levels of CRP, IL-6,IL-8, NF-κB increased significantly(P<0.05). In Shengwei Prescription group, hericium group, or Shengwei Prescription + hericium group, significant increased levels of PGII,PGI, GAS and EGF(P<0.01 for Shengwei Prescription + hericium group, P<0.05 for Shengwei Prescription or hericium group), and decreased levels of CRP, IL-6, IL-8 and NF-κB were detected in CAG rats (P<0.05 for each group).Conclusion:1、Experimental group and control group can effectively improve the clinical symptoms of patients, but the experimental group is better than the control group(P<0.01).2、The experimental group and the control group can both improve CAG patients in terms of gastric mucosa chronic inflammation, glaular atrophy and intestinal metaplasia. But the effect of the experimental group is better than control group.3、After the drug treatments, gastritis mucosal atrophy and intestinal metaplasia in chronic atrophic gastritis rats have different degrees of improvement, and Shengwei Prescription+hericium group’s effect is the best.4、The serum levels of PGI, PGII, GAS and EGF decreased in CAG groups, while the serum levels of CRP, IL-6, IL-8 and NF-κB increased.5、ShengweiPrescription, Hericium, and ShengweiPrescription+Hericium can improve the levels of PGI, PGII, GAS and EGF in the CAG rats, and decrease the level of CRP, IL-6, IL-8 and NF-κB. This may be the possible mechanism of Shengwei Prescription’s therapeutic effect for CAG patients.
