To explore the influence of earthworms on fat metabolism of lipid-lowering active ingredients, and experiment with guinea pigs as an object to study the lipid-lowering effect, provide the basis for the development of research earthworm extract.Methods:1.Preparation and separation of effective components earthworms:to clean earthworms were homogenized by sonication and then centrifuged to miscellaneous; by ammonium sulfate precipitation and ultrafiltration methods, interception earthworm protein 3kd to 100kd segment as earthworms lipid molecules effective preparation.2, Animal lipid-lowering trials:in 30 male guinea pigs weighing about 240g were randomly divided into five groups, each 6, respectively, the control group, high, medium and low-dose treatment group and positive control group, during the experiment in addition to the control group, the other groups were to build high-fat model, various lipids and liver function were measured after 4 weeks; liver paraffin tissue sections, and to observe, describe, and photomicrography.The results and analysis:1, Clinical manifestations:the experiment, most of the positive control group of guinea pigs show increased abdominal circumference, drowsiness, do not like moving; experimental guinea pigs have similar performance slightly. After the test load, the positive control group and the control group were significantly different compared to body weight (P<0.01); low-dose treatment group was significantly lighter weight than the positive control group (P <0.05), while in the high-dose treatment group weight significantly lighter than the positive control group (P0.01).2,Liver histopathological changes:visual observation gross specimen found that, compared with the control group, the positive control group of guinea pig liver volume was significantly larger, lighter color. Compared with the positive control group, the experimental group of low, medium and high dose guinea pig liver size and color gradually become normal; liver tissue section microscopy, the positive control group was observed in guinea pig liver cells have significant steatosis and hepatic cell boundaries is not obvious, low, medium and high doses of the experimental group guinea pig liver biopsy tissue gradually reduced steatosis, lobular and gradually returned to normal from the edge of the area to the central vein of liver cells.3, Fat metabolism and liver function status(1) Lipid parameters:Compared with the control group, the positive control group of guinea pigs of TC, TG, LDL-C were significantly increased (P<0.01), HDL-C levels were significantly lower (P<0.01); and the positive control group relatively low dose group of guinea pigs TC and dose experimental guinea pig TG, LDL-C and high-dose groups of guinea pigs TC, TG, LDL-C were highly significantly decreased (P<0.01), and HDL-C were significantly higher (P<0.05).(2) Liver function:Compared with the control group, the positive control group of guinea pigs AST and ALT were significantly increased (P<0.01); the other groups were not significantly different (P> 0.05).Conclusion:1, the active ingredient earthworm has obvious lipid-lowering effect, particularly on serum triglycerides and cholesterol have good regulation.2, earthworm extract has a protective effect on the liver, can significantly reduce hyperlipidemia, liver tissue damage; liver steatosis eased.
