Hyperlipidemia(HLP)is known as the key factor to many cardiovascular and cerebrovascular diseases.What's more,it can also give rise to a series of diseases that are harmful to people's health.the morbidity of the hyperlipidemia is increasing due to the rising number of elderly people.For the further study of hyperlipidemia is of great importance to find a way that can both regulate blood lipid and security.Traditional Chinese medicine(TCM)has a couple of ways that are valuable in preventing or treating diseases.Blood-pricking therapy is one of the traditional acupuncture method which has strong security,simple and the adverse reactions are mild and tolerable.It's proved that blood-pricking therapy can bring down blood viscosity,thus triggers the change of signal transduction in vascular smooth muscle cell(VSMC).Motivate the regulatory reaction in intracellular,extracellular,middle of the graft and the whole body.Relevant recordings convinced that hyperlipidemia can be treated by pricking out blood.This experiment is on the base of my mentor's clinicalexperience and related literature.We induced hyperlipidemia model by fed them with high fat diet.Them pricking out blood in Fenglong,Zusanli and somewhere near them and observe the change of blood lipid,vascular endothelial cell function and lipid metabolic enzyme.Finallywe can draw a conclusion to the underlying mechanism of blood-pricking therapy in treating hyperlipidemia,and translate the findings from fundamental research into clinical application.Objective:This experiment induced hyperlipidemia model by fed them with high fat diet.Them pricking out blood on Fenglong,Zusanli and somewhere near them and observe the change of blood lipid,vascular endothelial cell function(NO,ET-1)and lipid metabolic enzyme(LPL,HL).Finallywe can draw a conclusion to the effect and underlying mechanism of blood-pricking therapy in treating hyperlipidemia.Method:32 male SD rats were randomly divided into normal control group,model group,blood-pricking group and drug therapy group,8 cases each.Rats in normal control group were fed by standard diet,others were treated with high fat feed to conduct the hyperlipidemia model.Normal control group and model group received none treatment but grab and press after the HLP model established successfully.Blood-pricking group got hemorrhaged 0.3ml-0.5ml per time and 2 times a week for 4 weeks on Fenglong,Zusanli and somewhere near them.Drug therapy group got simvastatin2mg/kg/dfor 4 weeks.All the rats got open surgery to take blood under 4%chloral hydrate anesthesia.8 indexes were carried on the research,they are total serum cholesterol(TC),triglyceride(TG),HDL cholesterol(HDL-C),LDL cholesterol(LDL-C),nitric oxide(NO),endothelin-1(ET-1),lipoprotein lipase(LPL)and hepatic lipase(HL).Result:1.Rats in normal control group were generally in good condition.Model group's rats showed a spiritual burnout,slow in reaction,body odor and their for were losing lustrous.After treatment,general condition in blood-pricking group and drug therapy group obtain the varying degree alleviating,but not as good as normal control group.2.After the adaption for 1 week,there found no differences on weight in each group(P>0.05).Rat's weight showed a increasing in different degree from week 1 to 4.normal control group was the slowest one in increasing of body weight.Intervention measures were taken from week 5 to 8.Body weightof rats in blood-pricking group and drug therapy groupdiminished rapidly.As indicated by body weight in those 2 groups,It had a significantly higher value than normal control group and model group(P<0.05).What's more,body weight of rat's in model group also showed a downtrendfromweek 6.And it showed nostatistical significance(P>0.05).3.Compared with the control group,the level of TC?TG?LDL-C in the model group significantly increased,the level of HDL-C content in the model group significantly decreased,and the difference had a statistical significance(p<0.01).Compared with the model group,the levels of TC?TG content in the blood pricking group and medication group decreased,and the difference had a significant statistical significance(p<0.01).The levels of LDL-C content in the blood pricking group and medication group decreased,and the difference had a statistical significance(p<0.05).The levels of HDL-C content in the blood pricking group and medication group increased,and the difference had a statistical significance(P<0.05)4.Compared with the control group,the level of NO in the model group significantly decreased,the level of ET-1 content in the model group significantly increased,and the difference had a significant statistical significance(p<0.01);Compared with the model group,the levels of NO content in the blood pricking group increased,the levels of ET-1 content decreased,and the difference had a statistical significance(P<0.05),The levels of NO content in the medication group increased,the levels of ET-1 content decreased,and the difference had a significant statistical significance(P<0.01).5.Compared with the control group,the level of LPL in the model group significantly decreased,and the difference had a significant statistical significance(p<0.01).The level of HL in the model group significantly decreased,and the difference had a statistical significance(p<0.05).Compared with the model group,the levels of LPL?HL content in the blood pricking group and medication group increased,and the difference had a Statistical significance(P<0.05)Conclusion:1.It's showedthat blood-pricking on Fenglong and Zusanli has the ability to refrain the weight gain by a high-fat diet.Reduce the further developmentof HLP.2.Blood-pricking on Fenglong and Zusanli can depress the content of TC?TG?LDL-C and improve HDL-C in rat's serum.Meet the aim of regulate the lipid metabolism by suppressing excessive secretion of lipid and redress lipid metabolism disorders.3.Blood-pricking on Fenglong and Zusanli truly can increase NO and decrease ET-1.The mechanism might be regulate the systolic and diastolic function of blood vessels,delay and alleviate the injury degree of endothelial cells,thus to protect the endothelial cells.4.By blood-pricking on Fenglong and Zusanli,the activity of LPL?HL increased significantly.Those two indexes were activated to promote lipid metabolism and hydrolysis.Finallythe level of lipid metabolism can be corrected.
