ObjectiveThrough literature and animals experimental researchs,the study on the mechanism of Jiangzhuotiaozhi Fang regulating the lipid metabolism in hyperlipidemia rats based on PPAR-α pathway is discussed,which provides a reference for clinical research and development of new drugs.Methods1.Literature study:Summary of the modern medicinc and traditional Chinese medicine on hyperlipidemia understanding and treatment of related principles and expounds Professor Xian-ShaoXiang’ s thought of treating hyperlipidemia in clinical.2.Experimental study:Using high fat emulsion irrigation stomach method to duplicate the hyperlipidemia rat model and according to the treatment of the principle of administration,with di fferent dose of Jiangzhuotiaozhi Fang and positive control group method of intervention in rats,biochemical method to detect serum TG concentration;to observe the pathological change of aortic and PPAR-a and VCAM-1 immune group of positive expression;detection of body weight,wet weight of liver,liver coefficient and hepatic inflammation and steatosis degree,ELISA for the detection of hepatic LPL content,RT-PCR and Western blotting detection of PPAR-αmRNA and protein expression level.Results1.Literature research showed hyperl ipidemia occurred in diet,overwork,endowments and other factors,differentiation of symptoms and signs are yet to be reunified,the traditional Chinese medicine in the dual regulation of lipid metabolism has unique advantages.Professor Xian-ShaoXiang combined with years of clinical experience and literature analysis,and put forward the disease belongs to the virtual reality of the certificate,the key pathogenes is to temper weak for this,caused by phlegm and blood stasis standard.The main organs of the lesion in the spleen,kidney,liver,should be treated by Qi.2.The experimental study successfully replicated the model of hyperlipidemia rats.The serum concentration of TG can be decreased by Jiangzhuotiaozhi Fang,which can be significantly higher in middle and high dosage(P<0.05).Drug intervention group rat aortic intimal lesions significantly reduced,into the endometrial thickness and vacuolar also reduced;PPAR-α expression in the drug intervention group high fat model group increased,between each drug intervention group had no statistically significance(P>0.05).Compared with the normal control group,the expression of VCAM-1 in the high fat group was higher than that in the control group,but there was no significant difference between the two groups(P>0.05).The fatty liver disease of rats in each drug intervention group was improved(P>0.05).The expression level of LPL was significantly different between the middle dose of Jiangzhuotiaozhi Fang and the high fat model group(P<0.05).With high lipid model group,the rats liver PPAR-αmRNA and PPAR-α protein expression increased significantly,fenofibrate capsule and Jiangzhuotiaozhi Fang groups no significant difference(P>0.05).Conclusions1.Heoretical study shows that,Jiangzhuotiaozhi Fang with philosophy of compatibility of prescription and consistent and coherent of disease and syndrome research ideas,with spleen and kidney,dispersing stagnated liver qi and activating blood,regulating qi and resolving phlegm,detoxification reduced turbidity effect for Qi deficiency and phlegm and blood stasis type high lipid in patients with hyperlipidemia.2.Experimental studies show that Jiangzhuotiaozhi Fang can significantly reduce the high fat diet rats serum TG concentration levels,liver index,improve aortic inner and outer membrane and liver fatty change.Jiangzhuotiaozhi Fang by upregulating PPAR-α reduced the expression of VCAM-1,decrease the thickness of the intima,improve aortic fatty change is not clear.Jiangzhuotiaozhi Fang can be through upregulation of PPAR-α,promote LPL expression,decreased serum TG concentration,relieve liver fatty degeneration and to reduce turbidity,the effect of middle dose of Jiangzhuotiaozhi Fang is the best.
