Objective: To establish a quality control method of Arctium tomentosum Mill. seeds; To explore the antioxidant activity and inhibition of α-glucosidase activity of polysaccharide in Arctium tomentosum Mill. seeds; To study the monosaccharide composition and physicochemical properties of polysaccharides of Arctium tomentosum Mill. seeds. Methods: 1. Qualitative analysis of amino acids, at the same time qualitative and quantitative analysis of arctiin and arctigenin in Arctium tomentosum Mill. seeds by thin layer chromatography; Preliminary study of the chemical composition of Arctium tomentosum Mill seeds by infrared spectroscopy; The content of ten elements in the Arctium tomentosum Mill. seeds was determined by ICP-AES; 2. The antioxidant activities were examined from the aspects of reducing power, scavenging rate of DPPH, hydroxyl superoxide anion free radicals, and sod activity respectively. The hypoglycemic activity was examined by α-glucosidase inhibitory activities; 3. Adopt DEAE-cellulose column and Sephadex column separation purification of the active component of Arctium tomentosum Mill. seeds, Study on the active ingredient by using of UV, IR and TLC. Result: 1. The TLC isolated tryptophan, tyrosine, valine, serine, alanine and arginine. A TLC identification and TLCS method has been established for arctiin and arctigenin of Arctium tomentosum Mill. seeds. An optimized TLC method was used. The developer was consisted of chloroform and methanol(10:1), and under this condition arctiin and arctigenin can be completely separate, and spot color clear, the Rf value is moderate. The content of arctiin and arctigenin were 9.375%, 0.205%. Unique ingredients were got from different extracts by infrared spectroscopy analyze. The content of Cr, Cu, Fe, Mg, Mn, Ni, Pb, Sr, Ti, Zn was followed by 32.87, 20.2, 253.53, 3563.67, 26.34, 5.58, 2.89, 66.06, 3.27, 41.19μg·g-1. 2. The decreasing sequence of antioxidant activity was Vc> 20% ethanol elution> water elution fraction> 40% ethanol elution> 60% ethanol elution. The decreasing sequence of α-glucosidase inhibitory activities was acarbose> 20% ethanol elution> water elution fraction> 40% ethanol elution, 60% ethanol elution fraction was no inhibitory activity. 3. After DEAE-cellulose separted,four components W1, N1, N2, N3, obtain. Choose more components W1 purified by Sephadex G-100 column, and the elution curve was a single symmetrical peak. UV scanning spectrum showed purified W1’ has no obvious ultraviolet absorption peak at 260 nm and 280 nm, Infrared spectroscopy results show that arctiin polysaccharide has the general characteristics of polysaccharide absorption peak and containing absorption peak of alpha pyranoid sugar. Monosaccharide composition analysis showed that the separation and purification of polysaccharide W1’ was composed of glucose, arabia sugar xylose and galactose. Conclusion: 1. Provide a preliminary quality evaluation for the Arctium tomentosum Mill. seeds. 2. Crude polysaccharide 20% ethanol elution parts with good antioxidant activity and α-glucosidase inhibitory activity. 3. W1’ of the Arctium tomentosum Mill. seeds is a single polysaccharide group, does not contain nucleic acid and protein, belong to the alpha pyranoid sugar. Through thin layer chromatography preliminarily determines the monosaccharide composition of polysaccharide. This experiment provides reference basis for the comprehensive development and utilization of the polysaccharide and quality control system of Arctium tomentosum Mill. seeds.
