Cumin(Cuminum cyminum L.) fruit in peacekeeping medicine for the end of the grinding can be medicine for the treatment of cold stomach dyspepsia and abdominal pain. Cumin fruits main component consists of two parts: volatile and non-volatile components in the volatile components, more olefin compound, the main non-volatile components have more phenols, flavonoids, carbohydrates, lipids, proteins and other compounds. Flavonoids and polyphenols with a wide range of biological activity,such as anti-oxidation, anti-bacterial, anti-viral, anti-diabetes. In this paper, solvent extraction method to get cumin total phenol, total flavonoids, cumin and total phenol extraction process was optimized; while using different extraction section for the fruit Cumin organic solvent extraction system, with the corresponding evaluation method antioxidant(DPPH France, ABTS method, Hydroxyl radical process, ferricyanide reduction) of cumin total phenol, total flavonoids and antioxidant activity of different extracts to study at the same time by the enzymatic of cumin total phenol, total flavonoids and ?- glucosidase inhibitory properties were discussed.Cumin total phenolic content was measured using the Folin-Ciocalteu colorimetric determination of total flavonoid content cumin application of aluminum nitrate colorimetric method, cumin total phenol content determination process optimization using response surface methodology, preliminary purification of the total phenolic and total cumin flavonoids. The results are as follows:(1) Cumin total phenol extraction condition optimization results: 70% ethanol concentration, solid-liquid ratio and 60 ?, ultrasonic time, ultrasonic temperature 60 min. Under the condition of the process, cumin test content of total phenol was 13.812mg/g, higher predictive value of 4.5%. Macroporous resin method cumin total phenols preliminary purified crude extract, cumin total phenol content from 12.044 mg / m L up to about 20.186 mg / m L, display macroporous resin cumin total phenols have certain purification effect.From catechin equivalent purity 34% to 59%. Cumin flavonoids content: 27.7+1.3?g/m L; Alcohol soluble flavonoid content: 25.3+0.9?g/m L; Water soluble flavonoids content is 13.5+0.3?g/m L.(2) DPPH radical scavenging ability on the evaluation of the antioxidant strongest petroleum ether part: Vc > chloroform extracts of petroleum ether extract ethyl acetate extract>n-butyl alcohol extract ethanol extract purified total phenol > cumin total phenol > total flavonoids purified flavonoids; ABTS radical scavenging ability(concentration range 1 to 25 mg/m L) ethyl acetate parts of the strongest: Vc > n-buoh extract ethyl acetate extract purified flavonoids > total phenol chloroform extract purified petroleum ether extract > cumin total phenol > ethanol extract flavonoids;OH radical scavenging ability strongest ethyl acetate parts: Vc > n-buoh extract ethyl acetate extract of petroleum ether extract > total phenol > ethanol extract purified cumin > total phenol chloroform extract flavonoids > purified flavonoids; The total reducing power experiment, The strongest for cumin each extract total reducing power(concentration range 1 to 25 mg/m L) is n-butyl alcohol,These results can be separated in the future of antioxidative substances from certain data based of cumin.(3) the alpha- glycosidase enzyme inhibition experiment cumin extracts from different anti-diabetic activity, inhibitory activity of alpha glycosidase enzymes,cumin total phenol > positive control acarbose > cumin ethanol extracts > total flavonoids cumin cumin chloroform extracts > cumin petroleum ether extract cumin ethyl acetate extract > cumin n-butyl alcohol extract, cumin oil IC50 alue concentration of 72.41?L/m L. Enzymatic experiment results show that the different extracts from cumin has certain inhibition of alpha glycosidase enzymes, total phenol and cumin for alpha glycosidase enzyme inhibition effect is strongest, for cumin hypoglycemic activity to provide data for the further study of the reference.
