Objective: It is hard to accurately and completely control the intrinsic quality of Althaea rosea seeds, because there are only the names, sources, producing areas, characteristics and identifies in the Xinjiang Uygur Autonomous Region Uygur Medicine Standard(2010 Edition, Book 1). And this study aims to make a deeply and systematically studying of Althaea rosea seeds, to further expand the pharmacology effect of Althaea rosea seeds and to establish firm fundament of reasonable developing and utilizing to Althaea rosea seeds in Xinjiang.Method: The moisture, total ash and acid insoluble ash, water soluble extract and alcohol soluble extract of 10 batch Althaea rosea seeds were detected according to the Chinese Pharmacopoeia method. Total polyphenols, total triterpenes and total polysaccharides of 10 batch Althaea rosea seeds were determined by ultraviolet spectrophotometry. Artificial neural network model was applied to optimize polysaccharides from Althaea rosea seeds. The content of trace elements(Na, K, Mg, Ca, Cu, Mn, Fe, Zn, Cd, Pb, As and Hg) of Althaea rosea seeds were determined by atomic absorption spectrometry and atomic fluo-rescence spectrometry. Infrared spectroscopy regular pattern of 10 batch Althaea rosea seeds were explored by infrared spectroscopy and chemometrics. Study on the antioxidant activity of Althaea rosea seeds and its polysaccharide by using four kinds of antioxidant activity models(DPPH radical scavenging activity, Superoxide anion radical scavenging activity, Hydroxyl radical scavenging activity and Superoxide dismutase).Results: The moisture, total ash and acid insoluble ash, water soluble extract and alcohol soluble extract of 10 batch Althaea rosea seeds were 5.5%、14.75%、9.20%、23.15% and 12.43%, respectively. Total polyphenols, total triterpenes and total polysaccharides of 10 batch Althaea rosea seeds were ranged from 1.73-2.02、11.76-13.83 and 50.36-59.85 mg/g, respectively. A combination of an orthogonal L16(4)4 test design and a three-layer artificial neural network model was applied to optimize polysaccharides from Althaea rosea seeds extracted by hot water method. The highest optimal experimental yield of Althaea rosea seed polysaccharides of 59.85 mg/g was obtained using three extraction numbers, 113 min extraction time, 60.0% ethanol concentration, and 1:41 solid-liquid ratio. Under these optimized conditions, the Althaea rosea seed polysaccharides experimental yield was very close to the predicted yield of 60.07 mg/g and was higher than the orthogonal test results(40.86 mg/g). The content of trace elements(Na, K, Mg, Ca, Cu, Mn, Fe, Zn, Cd, Pb, As and Hg) of 10 batch Althaea rosea seeds were ranged from 44.15-167.68 、9604.52-18305.08 、 2378.37-3894.70 、 67.46-607.82 、 45.00-72.01 、 16.91-81.97 、442.09-2310.73、33.55-92.11、0.00-8.23、0.00-0.03、0.50-4.00 and 0.00-16.33 μg/g, respectively. The number of peaks, peak position, peak intensity, peak shape of 10 batch Althaea rosea seeds has some difference. The common peak was 728 cm-1, 1238 cm-1, 2854 cm-1, 2925 cm-1 and 3377 cm-1 through compared the infrared spectra of 10 batch Althaea rosea seeds and its extract. It can be used as characteristic peaks of the Althaea rosea seeds. Ten batch Althaea rosea seeds can be divided into three categories by using principal component analysis and cluster analysis method. The water extract and polysaccharide solution of Althaea rosea seeds have strong scavenging activity of DPPH free radical, hydroxyl radical and superoxide anion free radical, and contain a little superoxide dismutase.Conclusion: This research revised inspection item including moisture, total ash and acid insoluble ash, water soluble extract and alcohol soluble extract of Althaea rosea seeds. The content of total polyphenols, total triterpenes, total polysaccharides, trace elements of 10 batch Althaea rosea seeds were determined. The infrared spectroscopy regular pattern of 10 batch Althaea rosea seeds was developed. Meanwhile, the study on the antioxidant activity of Althaea rosea seeds and its polysaccharide found that it has good antioxidant effect. All of this offered the theory evidences for establishing scientific quality standard of Althaea rosea seeds.
