Background:Lung cancer is one of the most common cancer worldwide, and in the United States alone in 2013 there were 228190 new cases a year. Lung and bronchial cancer of cancer related death in 14% and 28% of new cancer cases. The past 10 years,in the developed countries to reduce smoking habits led the movement from smoking to reduce 1.3% of lung cancer cases each year. Developing countries, however,smoking has become more and more popular, the world and 1 billion more than 1billion male smokers and female smokers. Lung cancer prevention and control of the situation is not optimistic. Non-small cell lung cancer(NSCLC) accounts for about85% of lung cancer, including pathological on different adenocarcinoma and squamous cell carcinoma and large cell carcinoma subtype. If in the early stage diagnosis of non-small cell lung cancer, surgery is the most effective treatment method and its prognosis is good. Every time after the diagnosis of lung cancer, however, most patients(> 50%) is late.Lung cancer is the most common originated in the bronchial mucosa or gland malignant tumor, there is an upward trend in its morbidity and mortality, early diagnosis and treatment has important influence on the prognosis of patients with malignant tumor. And tumor markers earlier than imaging can reflect the existence of the tumor, progression or recurrence, tumor markers mainly refers to the produced by the tumor tissue or tumor arising from the interaction with the host can sign tumor presence of certain substances, from the human body is detected in the blood or other body fluids.Researchers from all over the world in recent years, the study of tumor is more and more in-depth, knowledge is becoming more and more comprehensive. And other malignant tumor, is the occurrence and prognosis of patients with lung cancer and the original oncogene activation and tumor-suppressor gene inactivation, hormone receptor levels, related to cell proliferation rate and so on many factors, especially the cell proliferation index, is a relationship to one of the important indices for the treatment and prognosis of patients with lung cancer. When the outside world a variety of carcinogenic factors acting on the human body, lead to the growth of the normal cells of the human body normal control out of control, the cells will appear from the regulation of proliferation. In tumor cell proliferation, DNA synthesis speed will increase sharply. Abnormal cell proliferation is the basis of tumorigenesis, observation can help to evaluate the nature of the tumor cell proliferation index, evaluation of prognosis.Thymidine kinase(TK) is the key enzyme in a pyrimidine relief way, catalytic terminal phosphate of ATP is transferred to 5 ’hydroxyl thymidine kinase(d Td R),production of deoxidization thymidylic acid(d TMP), then incorporated into DNA.There are two forms of thymidine kinase, cytoplasmic form of thymidine kinase 1(TK1), and in the form of a mitochondrial thymidine kinase 2(TK2). TK2 have independent of cell cycle and protein TK1 level closely related to cell cycle S- G2 phase, closely related to cell proliferation. TK1 throughout the cell cycle plays a adjust the role of thymidine pool in cells, involved in DNA repair process. Cytoplasm thymidine kinase 1(TK1) is a kind of kinase is closely related to cell proliferation,TK1 in tumor cell cycle G1 late near the end of the began to rise, until the G2- M early began to rise sharply, growing at least 10 ~ 20 times. Serum TK1 in cells, and a healthy weight is still little or undetectable. The proliferation of cells and healthy serum TK1 level is very low or undetectable, and a significant rise in cancer patients serum TK1 level, up to 2100 times.Objective: To study the serum thymidine kinase 1(serum thymidine kinase 1, STK1)expression and clinical significance in patients with lung cancer.Method: Using western blot enhanced chemiluminescence detection of 127 cases of lung cancer patients treated first STK1 concentration before treatment, and analyze the clinical characteristics, some chemotherapy of 59 patients with lung cancer in clinical observation for 3 months, the other 50 healthy physical examination as a control.Results: The lung cancer groups before treatment STK1 concentration(4.78 + /- 6.84 rho mol/L) is significantly higher than normal control group(1.15 + /- 0.52 rho mol/L),t = 5.935, P < 0.001; Compared with lung cancer three STK1 higher sensitivity: STK1(73.2%) > CYFRA21-1(59.8%) > CEA(46.5%) > NSE(37.0%), STK1 joint obviously improve the detection sensitivity and CYFRA21-1(91.3%), four joint detection sensitivity was 95.3%; STK1 expression and pathological types of lung cancer(chi-square = 10.626, P = 0.005), the number of lymph nodes(chi-square =4.060, P = 4.060), and whether there is a distant metastases(chi-square = 4.226, P =4.226) and clinical stage(chi-square = 3.991, P = 3.991); No correlation with other clinical pathological characteristics, P values > 0.05; Chemotherapy patients, 3 months of blood serum TK1 level significantly lower than before treatment, P < 0.05).Conclusion: TK1 is valuable in lung cancer tumor markers, expressing differences in different pathological types of lung cancer, and its sensitivity to the diagnosis of lung cancer is the CEA, CYFRA21-1, NSE.
