Objective To test the function of Zhitongxiaojietang in the hyperplasia of mammary gland tissue of rats,to verify the curative effect of Zhitongxiao-jietang,bying observing the changes of pathologyicl morphology of rat with hyperplasia of mammary gland,sex hormone receptors expression of rat with hyperplasia of mammary glands.And then discuss the functional mechanism of Zhitongxiaojietang curing mammary gland hyperplasia from the perspective on adjustment of sex hormone receptors expression.Methods Select 70 SPF-grade female SD rats and divide them at random into control group,model confirmed group,Zhitongxiaojietang groups of large,me-dium,and low doses,as well as Rupixiao positive control group.Then estab-lished animal model of mammary glands with the combination of estradiol ben-zoate and progesterone.After the success of the building,respectively for drug treatment, Zhitongxiaojietang groups according to the concentration of large, medium and low dose lavage,positive control group lavage mixed suspen-sion.The normal group and model group was given amount of distilled water to fill the stomach,for 30 consecutive days.Fasting can not help water 12 hours after the last treatment,by intraperitoneal injection of 10% chloral hydrate anesthesia for rats, and take the second of right on full breast,observate pathological morphological changes of the breast tissue under optical micros-cope.Immunohistochemical staining was used to observe the expression of estr-ogen receptor(ER),progesterone receptor(PR)and androgen receptor(AR) in rat mammary gland.At the same time,the content of the receptor was semi quantitati-ve analyzed.Results 1.Zhitongxiaojietang impact on the general condition of rats:compared with normal group rats,the model group there was a significantly increase in the nipple height and diameter of the second pairs of breasts(P<0.01).After giving a corresponding drug gavage treatment,each group rats nipple height and diameter than the model group significantly reduced(P<0.01).With Zh-itongxiaojietang group of large dose is particularly significant.Compared with Rupixiao group,the Zhitongxiaojietang low dose group had no significant diffe-rence.2.The effect of Zhitongxiaojietang on pathological morpholog-ical of rats:Normal group:Ductal epithelium of breast arrayedregularly.No expansion was in ductal cavity and acinar cavity.The number of lobule of ma-mmary gland and the gland alveolusin lobule of mammary gland were very few.Model group: Hyperplasia of mammary glands is diffuse.Hyperplasia of the Gland alveolus and ductus was in great number.Gland alveolus in lobule of mammary gland was intensive and inequality of size.Obviously,there was ex-pansion in ductal cavity and acinar cavity.And so much secretion was in intracavit-y.After treatment with drug lavage,low dose group of Zhitongxiaojietang does not change significantly, middle and high dose group compared with model gr-oup,the number of the Gland alveolus was few and the secretion was so lit-tle.Hyperplasia of the Gland alveolus in Rupixiao control group was in diffe-rent degree.However, expansion in ductal cavity and acinar cavity was not obvious.The secretion was little.3.ER,PR and AR expression quantity chang-es:rats compared with normal group,model group rats breast tissue in the ER,PR and AR expression significantly higher(P<0.01).After drug gavage tre-atment the pain subsides knot decoction high dose group can significantly reduce breast rights in breast tissue in rats of the expression of ER, PR and AR protein(P<0.05 or P<0.01).And better than that of breast elimination group(P<0.05).Conclusion Zhitongxiaojietang can reduce congestion,edema of the breast,im-proving mammary gland hyperplasia;Can adjust mammary glands ER, PR and AR protein expression,then treatment of rat mammary gland hyperplasia.
