Objective: To explore the relationship between the Candida albicans’ distribution in biofilm with early childhood caries(ECC) among 3~5 years old children of Uygur and Han nationality in Kashi Xinjiang, and the caries mechanism of Candida albicans in different genotypes. Method: The identified Candida albicans were genotyped by 25SrDNA-PCR technique; Tested the acid production, acid endurance and adhesive ability of Candida albicans; Compared the Sap vitality and the assay transcription level of SAP1, 2, 3, 4, 5 of C. albicans between the caries and caries-free(CF) groups of Uygur. Results: 1) The acquisition of C.albicans has a closely relationship with ECC.2) Genotype A is dominant in both Han and Uygur children who have caries. In Uyghur caries-free children’s biofilm, the C-type is the majority; 3) The acid production and acid endurance ability of C. albicans in Uyghur ECC are stronger than CF groups, the growth of ECC group are better than CF group too. When the PH value is 4.0 and 4.5, acid production and endurance ability of ECC group are stronger than CF group(P<0.05); 4) The averages ratio of ECC group glass wall adhesion are higher than the CF group(P=0.02); 5) All of Candida albicans strains showed Sap positive and the Sap level in ECC group are higher than the CF group(P=0.000; P=0.000). The RT-qPCRshowed the expression of SAP2 and SAP5 in ECC group was significantly higher than in the CF group(P=0.000, P=0.001); 6) The factorial analysis reveals that the three genotype groups was not statistically significant in different groups. When the glucose concentration was 0.1mol/L, the correlation between ECC and CF groups and genotyping were statistic meaningful(P=0.005), and the acid production ability of the three genotypes was statistically different(group P=0.020, genotype P=0.019), ECC group A>C>B, CF group B>A>C.The expression of SAP1, 2, 3, 4, 5 in the ECC and CF group, only SAP2 expressed different in genotypes between ECC and CF group(group P=0.001, Gene type P=0.020), SAP2 in the ECC group A>B>C; in the CF group B>A>C. Conclusion: 1) The ability of acid production and acid endurance of C. albicans, the ECC group are stronger than the CF; 2) The positivity of Sap and high expression of SAP2 and SAP5 may have close relation with ECC; 3) C. Albicans plays an important role in the progression of caries, and the genotype A carriers are more easier to suffer from caries. The cariogenic ability are different in different genotypes, this can be ascribe to the different environment vary from one person to another, but the specific mechanism still need further study.
